穿山甲、猪蹄甲、羊蹄甲炮制品水溶性成分的比较研究 被引量:3

摘要 目的:比较穿山甲、猪蹄甲、羊蹄甲炮制品的水溶性成分为寻找穿山甲替代资源提供参考。方法:本研究以穿山甲、猪蹄甲、羊蹄甲炮制品为研究对象,通过HPLC指纹图谱及多元数据分析对比3种炮制品的水溶性成分。结果:建立了穿山甲、猪蹄甲、羊蹄甲炮制品水溶性成分的指纹图谱,其中穿山甲炮制品指纹图谱确定了20个共有峰,并指认出7种成分,12批穿山甲炮制品相似度在0.813~0.944;猪蹄甲炮制品指纹图谱确定了22个共有峰,指认出4种成分,12批猪蹄甲炮制品相似度在0.882~0.991;羊蹄甲炮制品指纹图谱确定了19个共有峰,指认出4种成分,12批羊蹄甲炮制品相似度在0.865~0.998。穿山甲色谱峰可分为核苷类、肽类2类组分群;3种炮制品核苷组分相似,均含有黄嘌呤、次黄嘌呤、尿苷、尿嘧啶,L-甘-L-酪氨酸环二肽、L-丝-L-酪氨酸环二肽、D-丝-L-酪氨酸环二肽只存在于穿山甲中,3种炮制品的水溶性成分差异显著。结论:本研究系统比较了穿山甲、猪蹄甲、羊蹄甲、炮制品水溶性成分的异同,能够为其临床用药提供参考,对寻找穿山甲替代资源的具有参考意义。 Objective:In order to find alternative resources,the water-soluble components of processed products of pangolin scales,pig nail and sheep nail were studied in this study.Methods:The water-soluble components of three processed products were compared by HPLC fingerprint and multivariate data analysis.Results:The fingerprints of water-soluble components in processed products of pangolin,pig nail and sheep nail were established.Among them,the fingerprints of processed pangolin identified 20 common peaks and 7 components,the similarity of 12 batches of processed pangolin ranged from 0.813 to 0.944;the fingerprints of processed pig nail identified 22 common peaks,and 4 components.The similarity of 12 batches of processed pig nail was 0.882~0.991;meanwhile,19 common peaks and 4 components were identified in the fingerprint of processed sheep nail,and the similarity of processed sheep nail was 0.865~0.998.The chromatographic peaks of pangolin scales could be divided into two groups:nucleosides and peptides.The nucleoside components of the three processed products were similar and there were many common components.The nucleoside components of the three processed products were similar,including xanthine,hypoxanthine,uridine and uracil,but L-glycine-L-tyrosine cyclic dipeptide,L-silk-L-tyrosine cyclic dipeptide and D-silk-L-tyrosine cyclic dipeptide only existed in the processed pangolin.There were significant differences in water-soluble components among the three processed products,but the peptide components were significantly different.Conclusion:This study compares the similarities and differences among the processed products of pangolin,pig nail and sheep nail from the perspective of water solubility,which can provide reference for the clinical use of these three traditional Chinese medicine.
