摘要
藜麦是唯一一种单体植物即可满足人体基本营养需求的食物,其籽粒脂肪酸含量丰富、组成合理。植物脂肪酸转运蛋白FAX对介导质体中脂肪酸向外运输过程中发挥着重要的作用。本研究基于藜麦基因组序列信息,对藜麦FAX基因家族进行全基因组鉴定,并对该家族成员的基因结构、染色体定位、蛋白结构和表达模式等进行了全面分析,为进一步了解该家族功能和培育油用藜麦品种提供理论支撑。结果表明,藜麦FAX基因家族包含10个成员,分布在8条染色体上,8个CqFAX基因包含内含子和外显子;蛋白序列对比结果显示藜麦CqFAX蛋白之间氨基酸序列保守性较低,只有少数氨基酸残基具有保守性,可能对于FAX蛋白功能发挥关系密切;大多数CqFAX含有4个α螺旋,与其三级空间结构相似;进化树分析表明CqFAX蛋白分为4枝,与基因结构关系基本保持一致。对转录组表达数据分析表明,CqFAX基因在藜麦不同组织中均存在表达,其中AUR62006421、AUR62017596、AUR62026084和AUR62029770在种子中表达水平较高,可能在种子脂肪酸形成和积累中发挥着重要作用。
Quinoa is the only single plant that meets the basic nutritional needs of the human body.Its grain fatty acid content is rich,the composition is reasonable.Plant fatty acid transporter FAX plays an important role in mediating the outward transport of fatty acids from plastids.Based on the genome sequence of quinoa,the FAX gene family was identified,and the gene structure,chromosomal location,protein structure and expression pattern were analyzed comprehensively,which provided theoretical support for further understanding of the function of this family and cultivation of oil quinoa varieties.The Quinoa FAX gene family consisted of ten members,which were distributed on eight chromosomes.The gene structure result showed that eight CqFAXs contain intron and exon.Protein sequence alignment analysis showed that CqFAX protein sequence had a relatively low amino acid sequence conservation,which may play an important role for FAX protein.Most CqFAX proteins contained four transmembrane domains and had similar 3 D modellings.Based on phylogenetic tree analysis,the CqFAX protein was divided into 4 branches,which were basically consistent with the gene structure.The expression pattern of CqFAX family genes were analyzed by transcriptome sequencing,AUR62006421,AUR62017596,AUR62026084 and AUR62029770 were highly expressed in seed of quinoa.The results indicated that the four CqFAX genes were in accordance with the oil accumulation process.
