摘要 利用大孔树脂法分离纯化黑米花色苷。考察树脂类型、洗脱液乙醇浓度、上样液的浓度、洗脱液的流速等因素对分离纯化效果的影响,确定了大孔树脂法分离纯化黑米花色苷的工艺条件:选用D101大孔树脂、乙醇洗脱液的体积分数60%、上样液中花色苷的质量浓度12.0 mg/mL、上样液pH 3.0、洗脱速度3.5 mL/min、D101大孔树脂上样液中花色苷最大量7.2 mg/g。在此条件下,经大孔树脂分离纯化后所得纯化物中花色苷的纯度达到62%,证明利用D101大孔树脂分离黑米花色苷的方法可行。 Using macroporous resin to separate and purify the anthocyanins in black rice. The effects of two macroporous resins types, the elution liquid of ethanol concentration, the sample concentration, the elution speed, and other influence factors on purification were studied. The optimum separation conditions of anthocyanins in black rice using macroporous resin were as follows: type of macroporous resin was D101, the volume concentration of ethanol elution was 60 %, the sample concentration was 12. 0 mg/mL,pH was 3. 0, elution speed was 3. 5 mL/min, and the sample amount was 7. 2 mg/g macroporous resin.After separation and purification by macroporous resin with the optimum conditions, the purity of anthocyanins in extract was up to 62 %, which indicated that it was feasible to separate and purify black rice anthocyanins with D101 macroporous resin.
