摘要
目的:观察甘草附子汤对胶原诱导性关节炎(CIA)大鼠的治疗作用,初步探讨其对大鼠类风湿性关节炎的影响。方法:将60只Wistar大鼠随机分成正常组,模型组,甘草附子汤低、中、高剂量组,MTX组,每组10只,建立CIA模型,造模成功后,各组连续灌胃给药2周,1次/d,甘草附子汤低、中、高剂量组给药剂量分别为4、8、16g·kg^(-1)·d^(-1),MTX组给药剂量为1mg/kg,每周1次,正常组和模型组给予等体积的0.9%氯化钠溶液。观察大鼠足趾肿胀程度,采用ELISA试剂盒测定血清和关节液中白细胞介素(IL)-1β、IL-10及肿瘤坏死因子-α(TNF-α)含量。结果:与正常组比较,模型组大鼠足趾容积显著增加(P<0.05,P<0.01),血清及关节液中IL-1β、TNF-α水平显著升高(P<0.01,P<0.05),IL-10水平显著降低(P<0.05);与模型组比较,甘草附子汤中、高剂量组和MTX大鼠足趾容积显著减小(P<0.05),甘草附子汤高剂量组和MTX组血清和关节液中IL-1β、TNF-α含量显著降低(P<0.01,P<0.05),IL-10含量显著增加(P<0.05,P<0.01)。结论:甘草附子汤可能通过降低CIA大鼠血清和关节液中IL-1β和TNF-α含量、增加IL-10含量发挥抗炎作用,起到对CIA大鼠膝关节的保护作用。
Objective: To observe the therapeutic effect of Gancao Fuzi Decoction on collagen-induced arthritis in rats and to explore the effects on rheumatoid arthritis in rats. Methods: Sixty Wistar rats were randomly divided into six groups(normal group, model group, Gancao Fuzi Decoction low, middle and high dose groups and model group) with 10 rats in each group. Except the normal group, the rheumatoid arthritis model was established. After the success of the model, each group was intragastrically administered for 2 weeks, and the dosage of Gancao Fuzi Decoction in the low, middle and high dose groups was 4, 8, 16 g · kg^(-1)·d^(-1), respectively. The dose in model group was 1 mg/kg, once a week, and the normal group and the model group were given the same dose of normal saline. The paw volume of rats was tested, and the expression of IL-1β, IL-10 and TNF-α in serum and synovial fluid were measured by ELISA kit. Results: Compared with the control group, the paw volume in the model group increased significantly(P<0.05, P<0.01), and the levels of IL-1β and TNF-α in serum and synovial fluid increased significantly(P<0.01, P<0.05), the levels of IL-10 decreased significantly(P<0.05). Compared with the model group, the paw volume in the Gancao Fuzi Decoction middle dose group, high dose group and MTX group were significantly decreased(P<0.05), and levels of IL-1β and TNF-α in serum and synovial fluid were significantly decreased(P<0.01, P<0.05), the level of IL-10 was significantly increased in Gancao Fuzi Decoction high dose group and MTX group(P<0.05, P<0.01). Conclusion: Gancao Fuzi Decoction can protect the knee joint of rats with collagen-induced rheumatoid arthritis by inhibiting the levels of IL-1β, and TNF-α, increasing the level of IL-10 in serum and synovial fluid.
