大麦若叶青汁粉总三萜超声提取工艺研究 被引量:6

摘要 以大麦若叶青汁粉为原料,乙醇作为提取溶剂,采用超声波辅助提取总三萜,探究大麦若叶青汁粉提取总三萜的最优条件.从液料比、乙醇浓度、超声时间和超声功率4个方面进行单因素试验,以大麦若叶青汁粉总三萜的产率为响应值,利用响应面法进行产率预测和工艺分析,并加以验证.所得最优工艺条件为:乙醇79.97%、液料比为24.20∶1、提取22.50 min,大麦若叶青汁粉总三萜预测值为22.70 mg/g.根据实际调整方案为:乙醇浓度80%、液料比24∶1、超声22 min,大麦若叶青汁粉总三萜得率为21.69 mg/g,通过单样本t检验,实测值和预测值之间无显著差异(P>0.05),说明该方法可优化大麦若叶青汁粉总三萜的提取. In the present study,total triterpenes were extracted from barley young leaves using ultrasonic assisted extraction.The Response Surface Method was employed to optimize the extracting conditions,and three factors and four levels analysis were carried out based on single-factor test,including the solid-liquid ratio,ethanol concentration,ultrasonic time and ultrasonic power.The optimal conditions were 79.97%of ethanol concentration,24.20∶1 of liquid-solid ratio,22.50 min of extraction time,and predicted value of total triterpenes was 22.70 mg/g.According to the actual adjustment with an ethanol concentration of 80%,a liquid-solid ratio of 24∶1,and an ultrasonic time of 22 min,the extraction rate of total triterpenes was 21.69 mg/g.One-sample test was conducted and showed that no significant differences(P>0.05)were found between the actual yield and the predicted value,indicating that this method was suitable for the optimization of total triterpenes extraction from barley young leaves.
