摘要
为建立一套优化的组织培养和遗传转化再生体系,以加工番茄新品种新番72号为试材,分别选取4~8 d苗龄无菌苗的子叶和下胚轴为外植体,研究了不同苗龄、外植体、激素浓度配比对番茄愈伤组织诱导、不定芽分化的影响,以建立番茄再生体系,为下一步基于CRISPR-Cas9定点突变改良新疆加工番茄耐贮性奠定基础。结果表明,新番72号加工番茄再生体系建立中以苗龄6 d的子叶为最佳外植体,愈伤诱导、芽分化率及生长情况均优于下胚轴;愈伤诱导最佳培养基为MS+0.5 mg·L^(-1)IAA+2.0 mg·L^(-1)6-BA+2.0 mg·L^(-1)ZT,子叶的诱导率为90.5%,下胚轴的诱导率为80.8%;继代培养芽分化的最佳培养基为MS+0.2 mg·L^(-1)IAA+1.0 mg·L^(-1)6-BA+1.0 mg·L^(-1)ZT,子叶的芽分化率较高,可以达到85.3%。
To establish a set of optimal tissue culture and genetic transformation regeneration system,Xinfan 72 was used as material in this research,the cotyledon and hypocotyl of 4-8 days of aseptic seedlings were selected as explants,the effects of different seedling ages,explants and hormone concentrations on callus induction and adventitious bud differentiation in tomato were studied,in order to establish tomato regeneration system,lay the foundation for the next preliminary research on improving the storage tolerance of Xinjiang processed tomato based on CRISPR-Cas9 site-directed mutation.The results showed that cotyledons of 6d seedling age were the best explants in the regeneration system of Xinfan 72 tomato,and the callus induction,bud differentiation and growth were better than hypocotyl.The optimal callus induction medium was MS+0.5 mg·L^(-1) IAA+2.0 mg·L^(-1)6-BA+2.0 mg·L^(-1) ZT,and the induction rate of cotyledons and hypocotyls were 90.5%and 80.8%,respectively.The best medium for bud differentiation in the secondary culture was MS+0.2 mg·L^(-1) IAA+1.0 mg·L^(-1)6-BA+1.0 mg·L^(-1) ZT.
