摘要
目的研究miR-9-5p、miR-125b-5p在膝关节骨性关节炎(KOA)软骨中的表达及意义。方法选取骨科建卡就诊的KOA患者124例为研究对象(KOA组),将KOA组软骨标本(重度退变软骨、中度退变软骨、轻度退变软骨)分为重度组(32例)、中度组(44例)和轻度组(48例);选取同期因交叉韧带断裂、盘状软骨损伤等在医院建卡就诊的膝关节软骨无病变患者124例为对照组(NC组),通过实时荧光定量PCR(qRT-PCR)检测各组软骨组织中miR-9-5p、miR-125b-5p的表达水平,采用酶联免疫法检测各软骨组织中白细胞介素(IL)-1、IL-6、IL-8、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-13(MMP-13)、软骨寡聚基质蛋白(COMP)表达水平;Pearson相关分析KOA患者膝关节软骨中miR-9-5p、miR-125b-5p水平与IL-1、IL-6、IL-8、TNF-α、MMP-13、IL-1、MMP-13、COMP水平的相关性。结果KOA组患者软骨中miR-9-5p表达水平相对于NC组显著下调(P<0.05),miR-125b-5p表达水平显著上调(P<0.05);重度组、中度组、轻度组miR-9-5p表达水平依次升高(P<0.05);重度组、中度组、轻度组miR-125b-5p表达水平依次降低(P<0.05);轻度组、中度组、重度组KOA患者软骨中IL-1、IL-6、IL-8、TNF-α、MMP-13、COMP表达水平依次升高(P<0.05);KOA患者软骨miR-9-5p水平与IL-1、IL-6、IL-8、TNF-α、MMP-13、COMP、miR-125b-5p水平呈负相关(P<0.05);miR-125b-5p水平与IL-1、IL-6、IL-8、TNF-α、MMP-13、COMP水平呈正相关(P<0.05)。结论miR-9-5p在KOA患者膝关节软骨组织中表达下调,miR-125b-5p表达上调,两者可能通过影响炎症因子水平,参与KOA疾病发生、发展。
Objective To study the expressions and significances of miR-9-5 p and miR-125-5 p in cartilage of patients with knee osteoarthritis(KOA).Methods A total of 124 KOA patients who visited the orthopedics in our hospital from January 2015 to January 2019 were selected as study group(KOA group),124 cases of patients without KOA visited our hospital due to cruciate ligament rupture and discoid cartilage injury were selected as control group(NC group).The expression levels of miR-9-5 p and miR-125 b-5 p in cartilage tissues were detected by real-time fluorescence quantitative PCR(qRT-PCR),the expression levels of IL-1,IL-6,IL-8,TNF-α,MMP-13,IL-1,MMP-13 and COMP in cartilage tissues were detected by ELISA.Pearson correlation analysis was used to analyze the correlations between the levels of miR-9-5 p,miR-125-5 p and the levels of IL-1,IL-6,IL-8,TNF-α,MMP-13,IL-1,MMP-13 and COMP in knee cartilage of KOA patients.Results The expression of miR-9-5 p in cartilage of KOA group was significantly lower than that of NC group(P<0.05),and the expression of miR-125 b-5 p was significantly higher than that of NC group(P<0.05).The expression of miR-9-5 p in severe group,moderate group and mild group decreased in turn(P<0.05).The expression level of miR-125 b-5 p in severe group,moderate group and mild group increased in turn(P<0.05).The expression levels of IL-1,IL-6,IL-8,TNF-α,MMP-13,IL-1,MMP-13 and COMP in cartilage of KOA patients in mild,moderate and severe groups increased in turn(P<0.05).The level of miR-9-5 p in cartilage of KOA patients was negatively correlated with the levels of IL-1,IL-6,IL-8,TNF-α,MMP-13,IL-1,MMP-13,COMP and miR-125 b-5 p(P<0.05).The level of miR-125 b-5 p was positively correlated with the levels of IL-1,IL-6,IL-8,TNF-α,MMP-13,IL-1,MMP-13 and COMP(P<0.05).Conclusion The expression of miR-9-5 p is down-regulated in knee cartilage tissue of KOA patients,and the expression of miR-125 b-5 p is up-regulated.Both of them may participate in the occurrence and development of KOA disease by affecting the level of inflammatory factors.
