摘要
以CuCl_(2)为原料,聚乙烯吡咯烷酮(K30)为模板,2-巯基苯并噻唑(MBT)为保护剂,用水合肼作还原剂,在常温下放置10 min,合成发射波长为606 nm的橙红色铜纳米粒子(CuNPs),用紫外、荧光和电镜对其光学性质和形貌进行表征。实验发现,在p H 8.3的Tris-HCl缓冲溶液中,盐酸小檗碱(Be-H)对CuNPs荧光强度有明显的猝灭作用,因此建立了一种以CuNPs为探针检测Be-H含量的新方法,实验考察了反应温度、时间、pH、缓冲溶液种类和用量对反应体系的影响,结果表明:Be-H含量在5.0×10^(-8)~6.0×10^(-5)mol/L范围猝灭程度ΔF与Be-H浓度呈线性关系,线性方程为ΔF=15.19c+60.02,R^(2)=0.9959,检出限为8.0 nmol/L,相对标准偏差为4.1%,该方法用于不同厂家盐酸小檗碱片中盐酸小檗碱的测定,回收率为99.4%~108.3%。
Copper nanocluster dots (CuNPs) with orange-red fluorescence at 606 nm was synthesized using CuCl_(2) as the raw material,polyvinyl pyrrolidone (K30) as the template,2-mercaptobenzothiazole as the stabilizer and hydrazine hydrate as the reducing agent at room temperature for 10 min.The luminescence characteristics of CuNPs were studied by fluorescence spectroscopy,ultraviolet-visible spectroscopy,and the morphology was studied by TEM.A novel method for quantitative determination trace berberine hydrochloride(Be-H) was developed based on the fluorescence quenching of CuNPs as probe in p H 8.3 Tris-HCl buffer solution.A wide range of reaction parameters,such as reaction temperature,reaction time,sorts and amount of buffer were examined.The results showed that theΔF was linearly related with Be-H concentration in 5.0×10^(-8)-6.0×10^(-5)mol/L.The linear equation wasΔF=15.19c+60.02 with correlation coefficient (R^(2)) of0.9959.The detection limit was 8.0 nmol/L,and the relative standard deviation was 4.1%.The method was used in the determination of Be-H content in different berberine hydrochloride tablets,and the recoveries were99.4%-108.3%.
