摘要
为阐明白纹雏蝗Chorthippus albonemus抗寒和高温耐受能力及其抵御机制,本研究测定白纹雏蝗过冷却点、冰点值及其对高温的耐受力,并采用生理生化方法测定白纹雏蝗在24、28、32、36、40、44和48℃不同温度下抗逆物质的积累量和变化情况。结果表明,白纹雏蝗卵的过冷却点值为-22.58℃,冰点值为-30.20℃。随着温度上升,白纹雏蝗的死亡率增加,在24~36℃温度范围,雌、雄成虫的死亡率均低于20.00%;44℃时,雌、雄成虫的死亡率分别为66.67%和63.33%,当温度上升至48℃时,雌、雄成虫的死亡率均达100.00%。白纹雏蝗雌、雄成虫体内的海藻糖含量均随着温度上升呈先上升后下降的波动趋势,不同温度处理间差异显著;36℃时,雌、雄成虫体内海藻糖含量均达到最大值,分别为0.195μg/g和0.197μg/g。白纹雏蝗雄虫的自由水与结合水的比值较雌成虫偏高,36℃时,雌、雄成虫比值均达到最大值,分别为35.10和84.70。在不同温度下,白纹雏蝗雌、雄成虫体内的抗逆物质油酸甲酯、亚油酸甲酯和亚麻酸甲酯3种不饱和脂肪酸含量不同,其中,雌成虫体内油酸甲酯、亚油酸甲酯和亚麻酸甲酯含量均在32℃达到最高,分别为74.18、51.62和117.35 mg/g,雄成虫体内油酸甲酯、亚油酸甲酯和亚麻酸甲酯含量分别在32、36和28℃温度下达到最大值,分别为30.19、39.38和104.57 mg/g,不同温度下雌、雄成虫体内3种不饱和脂肪酸含量均差异显著。说明白纹雏蝗具有较好的抗寒能力,并通过积累体内海藻糖含量增加呼吸代谢消耗的底物和调节自身体内脂肪酸的含量防止因水分过度蒸发抵御高温环境。
In order to clarify the cold resistance,high temperature tolerance and resistance mechanism of grasshopper Chorthippus albonemus,this study determined the supercooling point,freezing point and tolerance to high temperature of the grasshopper.Meanwhile,we measured the accumulation and changes of stress-resistant substances of the grasshopper under 24,28,32,36,40,44,48℃using physiological and biochemical methods.The results showed that the supercooling point and freezing point values were-22.58℃and-30.20℃,respectively.As the temperature rose,the mortality of C.albonemus increased.The mortality of male and female was within 20.00%in the temperature range of 24-36℃and66.67%and 63.33%,respectively,at 44℃.When the temperature rose to 48℃,the death rates of males and females were up to 100.00%.The trehalose contents in C.albonemus female and male fluctuated firstly and then decreased with rising temperature,and there were significant differences among different temperature gradients.The trehalose contents in both female and male reached the maximum(0.195μg/g and 0.197μg/g,respectively)at 36℃.The ratio between free water and bound water in C.albonemus male was higher than that in female,and both reached the maximum(35.10 and 84.70)at 36℃.Under different temperatures,the contents of stress-resistant substances,unsaturated fatty acids(including methyl oleate,methyl linoleate and methyl linolenate)in the bodies of female and male grasshoppers were different.The contents of methyl oleate,methyl linoleate and methyl linolenic acid in females all reached the highest at 32℃,which were 74.18,51.62 and 117.35 mg/g,respectively.The contents of methyl oleate,methyl linoleate and methyl linolenic acid in males reached their maximum values at three different temperatures(32,36 and 28℃),which were 30.19,39.38 and 104.57 mg/g,respectively.There were significant differences in the contents of three unsaturated fatty acids in females and males under different temperatures.The results indicated that C.albonemus had better cold resistance ability and resisted the high-temperature environment by accumulating the content of trehalose in the body to increase the substrate for respiratory metabolism and consumption and adjusting the content of fatty acids in the body to prevent excessive evaporation of water.
