马鞭草苷通过NF-κB/MAPK信号通路干预支气管哮喘大鼠气道炎症、气道平滑肌细胞增殖及RhoA表达的研究 被引量:6

摘要 目的:探究马鞭草苷对支气管哮喘大鼠气道炎症、气道平滑肌细胞(ASMCs)增殖、Rho A表达及NF-κB/MAPK信号通路的影响。方法:72只健康雄性SD大鼠随机分为对照组、模型组、地塞米松组和马鞭草苷高、中、低剂量组,每组12只。除对照组外,其余大鼠采用卵清白蛋白致敏法制备支气管哮喘模型,各组大鼠于模型制备成功次日开始接受药物灌胃治疗。记录各组大鼠支气管灌洗液中炎症细胞种类及计数情况,HE染色观察各组大鼠肺组织病理学变化,采用流式细胞仪检测细胞周期,采用PCR与Western blotting法检测大鼠肺组织Rho A表达水平,并分析马鞭草苷的作用机制。结果:与对照组比较,模型组大鼠支气管灌洗液中巨噬细胞水平明显降低(P<0.05),淋巴细胞、中性粒细胞、白细胞水平明显升高(P<0.05);S期细胞占比明显升高(P<0.05);Rho A表达水平明显上调(P<0.05);NF-κB/MAPK信号通路活性明显增加(P<0.05)。与模型组比较,马鞭草苷各剂量组和地塞米松组大鼠支气管灌洗液中炎症细胞计数明显减少(P<0.05),巨噬细胞水平明显增加(P<0.05);Rho A表达水平明显下降(P<0.05);NF-κB/MAPK信号通路活性明显降低(P<0.05)。结论:马鞭草苷可以减轻支气管哮喘大鼠气道炎症,抑制气道平滑肌细胞过度增殖,降低Rho A基因与蛋白表达水平,其具体机制可能涉及抑制NF-κB/MAPK信号通路活性。 Objective:To explore the effects of verbenalin on airway inflammation,airway smooth muscle cell(ASMCs)proliferation,Rho A expression and NF-κB/MAPK signaling pathway in rats with bronchial asthma.Methods:A total of 72 healthy male SD rats were randomly divided into control group,model group,dexamethasone group and verbenalin high,medium and low-dose groups,with 12 rats in each group.Except for the control group,the other rats were treated with ovalbumin sensitization method to prepare bronchial asthma models.The rats in each group began to receive drug gavage treatment the next day after the model was successfully prepared.The types and counts of inflammatory cells in the bronchial lavage fluid of each group were recorded.The pathological changes of the lung tissue of each group were observed by HE staining.The cell cycle was detected by flow cytometry.The expression of Rho A in lung tissue was detected by PCR and Western blotting,and the mechanism of verbenalin was analyzed.Results:Compared with the control group,the level of macrophages in the bronchial lavage fluid of the model group was significantly reduced(P<0.05),the levels of lymphocytes,neutrophils,and white blood cells were significantly increased(P<0.05),the proportion of S-phase cells was significantly increased(P<0.05),the expression level of Rho A was significantly increased(P<0.05),and the activity of NF-κB/MAPK signaling pathway was significantly increased(P<0.05).Compared with the model group,the number of inflammatory cells in the bronchial lavage fluid of the verbenalin high,medium and lowdose group and the dexamethasone group were significantly decreased(P<0.05),the level of macrophages was significantly increased(P<0.05),the expression level of Rho A was significant decrease(P<0.05),and the activity of NF-κB/MAPK signaling pathway was significantly decreased(P<0.05).Conclusion:Verbenalin can effectively reduce airway inflammation in asthmatic rats,inhibit the excessive proliferation of airway smooth muscle cells,and reduce the expression levels of Rho A gene and protein.The specific mechanism may involve inhibiting the activity of NF-κB/MAPK signaling pathway.
