摘要
目的探讨艾灸疗法对脑缺血大鼠海马CA1区的细胞损伤及海马谷氨酸转运体表达水平的影响,探讨其可能作用机制。方法雄性SD大鼠采用改良四血管阻塞法制备全脑缺血模型后分组:模型组、假手术组、艾灸组、艾灸+假手术组,每组10只。艾灸组和艾灸+假手术组予艾灸百会穴治疗。采用HE染色观察脑组织病理改变;采用蛋白免疫印迹法检测海马组织中胶质谷氨酸转运体-1(GLT-1)蛋白的表达;采用RT-PCR法检测海马组织GLT-1 mRNA表达。结果①与模型组相比,艾灸组海马CA1区锥体细胞损伤不明显,其组织细胞形态明显好转,组织学分级明显降低(P<0.05),神经元密度显著增高(P<0.05)。②艾灸疗法可上调脑缺血大鼠海马区GLT-1mRNA的表达(P<0.01)。③艾灸疗法可上调脑缺血大鼠海马区GLT-1蛋白表达(P<0.05)。结论艾灸疗法可促进脑缺血损伤海马区椎体细胞修复,其作用机制可能通过调控GLT-1的表达,调节神经递质和保护海马组织相关。
Objective To investigate the effect of moxibustion on cell damage and hippocampal glutamate transporter expression in the hippocampal CA1 region of cerebral ischemia rats,and to explore its possible mechanism. Methods Male SD rats were prepared by modified four-vessel occlusion method and then divided into four groups:model group,sham operation group,moxibustion group,moxibustion + sham operation group,10 rats in each group. Moxibustion group and moxibustion + sham operation group were treated with moxibustion Baihui acupoint. Pathological changes in brain tissue were observed with HE staining;Detection of GLT-1 protein expression in hippocampal tissue by Western blot;RT-PCR was used to detect the expression of GLT-1 mRNA in hippocampus. Results Compared with the model group,the hippocampal CA1 area of pyramidal cells in the moxibustion group was not significantly damaged,the histological cell shape was significantly improved,the histological grade was significantly reduced(P<0.05),and the neuron density was significantly increased(P<0.05). Moxibustion can up-regulate the expression of GLT-1 mRNA in hippocampus of cerebral ischemia rats(P<0.01). Moxibustion can increase GLT-1 protein expression in hippocampus of cerebral ischemia rats(P<0.05). Conclusion Moxibustion therapy can promote hippocampal vertebral cell repair in cerebral ischemic injury. Its mechanism may be related to regulating GLT-1 expression,regulating neurotransmitters and protecting hippocampal tissues.
