摘要
目的:探讨大黄对脑出血(ICH)大鼠的脑保护作用及作用机制。方法:雄性Wistar大鼠随机分为空白对照组、假手术组、ICH模型组、安宫牛黄丸组(0.27 g/kg)、大黄组(3.60 g/kg)。ICH模型组、安宫牛黄丸组和大黄组采用胶原酶注射尾状核法复制大鼠ICH模型。建模结束后安宫牛黄丸组和大黄组分别给予相应的药物灌肠(1次/d)。在给药后12 h、24 h、3 d、7 d分别取样,采用改良的神经功能损伤程度评分进行各组大鼠脑损伤程度评价;依文思蓝(EB)法检测各组大鼠血脑屏障的通透性;ELISA法检测各组大鼠血清中TNF-α、IL-6、IFN-γ、IL-4、IL-10、GSH含量;Western blot检测各组大鼠出血侧脑组织中Nrf2蛋白的表达。结果:与假手术组比较,ICH组EB含量和mNSS评分升高,血清中TNF-α、IL-6、IFN-γ含量均增加,IL-4、IL-10含量在出血后12 h升高,然后随着出血的进展随之降低,血清中GSH含量降低,出血侧脑组织中Nrf2蛋白的表达升高(P均<0.05);与ICH模型组比较,大黄组可以降低ICH模型大鼠的mNSS评分、脑组织的EB含量和大鼠血清中致炎因子TNF-α、IL-6、IFN-γ水平,提高抑炎因子IL-4、IL-10水平和大鼠血清中抗氧化因子GSH的含量以及出血侧脑组织中Nrf2蛋白的表达水平(P均<0.05)。结论:大黄通过抗炎和抗氧化作用保护ICH模型大鼠的血脑屏障,从而发挥脑保护作用。
Aim:To investigate the protective effects of Rhubarb on brain intracerebral hemorrhage(ICH)in rats and its mechanism.Methods:Male Wistar rats were allocated into blank control group,sham operation group,ICH model group,Angong Niuhuang pill group(0.27 g/kg),and Rhubarb extract group(3.60 g/kg).ICH model group,Angong Niuhuang pills group and Rhubarb extract group were performed to induce ICH rat model by collagenase injection via caudate nucleus.Angong Niuhuang pills group and Rhubarb extract group were given corresponding drug by enema,respectively,once a day after modeling.At 12 hours,24 hours,3 days,7 days after administration,the samples were taken,the degree of brain injury in rats was evaluated by improved neurological function injury score.The permeability of blood-brain barrier in rats of each group was detected by Evans blue method.The contents of TNF-α,IL-6,IFN-γ,IL-4,IL-10 and GSH in rat serum were detected by ELISA,and the expression of Nrf2 protein in the hemorrhagic brain tissue of the rats was detected by Western blot.Results:Compared with the sham operated group,the EB contents and mNSS scores in the ICH group were increased,the contents of TNF-α,IL-6,IFN-γin the serum were increased,the contents of IL-4,IL-10 were increased at 12 hours after ICH,and then decreased with the progress of hemorrhage,the content of GSH in serum was decreased,and the expression of Nrf2 protein was increased in the hemorrhagic brain tissue of ICH rats;compared with the ICH group,the mNSS score,EB content in brain tissue,and the inflammatory factors TNF-α,IL-6 and IFN-γlevels significantly reduced,the anti-inflammatory factors IL-4 and IL-10 levels and GSH in rat serum,as well as the expression of Nrf2 in Rhubarb extract group significantly increased(P<0.05).Conclusion:Rhubarb protects the blood-brain barrier of ICH model rats through its anti-inflammatory and antioxidant effects.
