摘要
目的:运用网络药理学方法研究桃仁-红花治疗高脂血症的作用靶点和信号通路,进一步分析其治疗高脂血症的物质基础和作用机制。方法:通过Drugbank、TTD数据库收集高脂血症作用靶点;通过中药系统学药理分析平台获得桃仁-红花所含化学成分及预测靶点;运用Cytoscape软件绘制“桃仁-红花活性成分-潜在靶点”网络;应用ClueGo对潜在靶点进行GO和KEGG富集分析。结果:桃仁-红花治疗高脂血症活性成分的潜在靶点网络中包含13个活性成分和16个潜在靶点,关键靶点涉及半胱氨酸天冬氨酸蛋白酶3、过氧化物酶体增殖物激活受体-γ、钠依赖去甲肾上腺素转运体、钠依赖5-羟色胺转运体、细胞色素P450、肿瘤坏死因子、胰岛素受体等;靶点基因功能偏于多巴胺、儿茶酚胺及含儿茶酚胺化合物的分解、甘油三酸酯隔绝、有机羟基化合物分解代谢等;KEGG通路涉及PPAR信号通路、药物代谢、可卡因成瘾、安非他明成瘾。结论:桃仁-红花通过增加靶基因转录、蛋白水解、神经递质转运的方式干预高脂血症的发生发展,其主要作用机制与药物代谢、PPAR信号通路相关。
Objective:To use network pharmacology to study the target and signal pathway of Taoren(Semen Persicae)-Honghua(Flos Carthami)n the treatment of hyperlipidemia,and to further analyze the material basis and mechanism in the treatment of hyperlipidemia.Methods:The target points of hyperlipidemia were collected by Drugbank and TTD(therapetic target datebase);the chemical components and prediction targets of Taoren-Honghua were obtained by TCMSP;the network of"Taoren-Honghua active components-potential target"was drawn by using Cyberscape;the potential target points were analyzed for GO and KEGG.Results:There were 13 active components and 16 potential targets in the network of active components-potential targets of Taoren-Honghua in the treatment of hyperlipidemia.The key targets included Caspase-3,peroxisome proliferator activated receptor-γ,sodium dependent norepinephrine transporter,sodium dependent 5-serotonin transporter,cytochrome P450,tumor necrosis factor and insulin receptor,etc.;the function of target genes was partial to the decomposition of dopamine,catecholamine and catecholamine containing compounds,triglyceride isolation,organic hydroxyl compounds catabolism,etc;the important KEGG pathway involved PPAR signaling pathway,drug metabolism,cocaine addiction and amphetamine addiction.Conclusion:Taoren-Honghua can interfere with the development of hyperlipidemia by increasing target gene transcription,protein hydrolysis and neurotransmitter transport.The main mechanism of the action is related to drug metabolism and PPAR signaling pathway.
