摘要
目的用网络药理学方法研究牛膝防治慢性牙周炎(chronic periodontitis, CP)的主要活性成分,以及分析其潜在分子机制和药理活性。方法在TCMSP数据库获取牛膝的主要化学成分及作用靶点,从Genecards等数据库搜索CP疾病靶点,通过韦恩分析获得交集靶点。构建牛膝防治CP的"成分-靶点"网络,并利用STRING数据库构建蛋白互作网络(protein protein interaction network,PPI network)。进行基因本体(gene ontology, GO)和京都基因和基因组百科全书(kyoto encyclopedia of genes and genomes, KEGG)富集分析后,构建"成分-靶点-通路"网络。结果获得牛膝活性成分20个,交集靶点183个,其中关键靶点包括丝氨酸/苏氨酸蛋白激酶1(AKT1)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF)等。牛膝治疗CP的主要活性成分有槲皮素、黄芩素、黄芩苷等。GO富集分析得到抑菌活性等952个GO条目。KEGG富集分析得到IL-17信号通路等101条通路。结论阐述了牛膝中治疗CP的主要成分、作用靶点和途径,为进一步开发牛膝中的主要成分来防治CP具有重要的研究价值。
Objective To study the main active components of Achyranthis Bidentatae Radix in the prevention and treatment of chronic periodontitis(CP)by network pharmacology,and analyze its potential molecular mechanism and pharmacological activity.Methods The main chemical constituents and targets of Achyranthis Bidentatae Radix were obtained from TCMSP database,the disease targets of CP were searched from Genecards and other databases,and the intersection targets were obtained by Venn analysis.The"component-target"network of Achyranthis Bidentatae Radix for the prevention and treatment of CP was constructed,and the protein-protein interaction network(PPI network)was constructed by using STRING database.After enrichment analysis of gene ontology(GO)and Jingdu gene and genome encyclopedia(KEGG),the"component-target-pathway"network was constructed.Results Twenty active components and 183 intersection targets of Achyranthis Bidentatae Radix were obtained,of which the key targets included AKT1,IL-6,TNF and etc.The main active components of Achyranthis Bidentatae Radix in the treatment of CP were quercetin,baicalein,baicalin and etc.A total of 952 GO items such as antibacterial activity were obtained by GO enrichment analysis.A total of 101 pathways such as IL-17 signal pathway were obtained by KEGG enrichment analysis.Conclusions The main components,targets and approaches in the treatment of CP in Achyranthis Bidentatae Radix are expounded,which has important research value for further development of the main components in Achyranthis Bidentatae Radix to prevent and treat CP.
