摘要
目的探究影响盐肤木叶中药用成分的因素。方法本研究对盐肤木叶多酚的提取工艺进行了优化,采用福林法测定了不同采摘时间和不同老嫩程度的盐肤木叶片的总酚含量,并采用超高效液相色谱测定了不同老嫩程度的盐肤木叶片中的6种成分的含量。结果最优的工艺条件为料液比1∶100(g/mL),提取液80%甲醇,超声时间50 min;在5月中旬至9月中旬,不同植株盐肤木叶片的多酚含量在同一月份内无显著差异(P>0.05);而盐肤木叶片的7月采摘时间对其多酚含量集与其他采集时间存在显著差异影响(P<0.05),并在7至8月含量达到峰值为(11.45±0.78)%,之后呈下降趋势。表没食子儿茶素没食子酸酯、表儿茶素没食子酸酯和根皮苷的含量均随盐肤木叶龄的增加而降低,并且在嫩叶和老叶间存在显著差异(P<0.05),根皮苷含量随树龄增加而增加,而没食子乙酯随树龄增加而减少,并且3年期树跟1年期树所产叶2种成分存在显著差异(P<0.05)。结论7至8月盐肤木叶片的总酚积累量达到峰值;与中老叶相比,嫩叶中的表没食子儿茶素没食子酸酯、表儿茶素没食子酸酯和根皮苷的含量较高,与1年期树叶片比,3年期树叶片中的没食子乙酯含量较低而根皮苷含量更高。
Objective To explore the main factors influencing the medical ingredients in the leaves of Rhus chinensis(R.chinensis).Methods The optimization of polyphenols extraction from the leaves of R.chinensis was explored and the change of components contents in R.chinensis leaves at different ages and different months was investigated by Folin method.Moreover,the contents of 6 components in R.chinensis leaves at different ages were determined by ultra-high performance liquid chromatography.Results The optimum extraction conditions were as follows:the methanol concentration was 80%,and the ratio of liquid to raw material was 1∶100(g/ml)with 50 min for extraction.There was no significant difference of the contents of polyphenols in R.chinensis leaves among different trees within the same month,whereas there was a significant difference in different months.Moreover,the polyphenols contents in leaves of R.chinensis increased with time first and peaked at(11.45±0.78)%in July and August and then decreased.The contents of epigallocatechin gallate,epicatechin gallate and phlorizin decreased with leaf aging,and there were significant differences between young and old leaves.With tree-age,the contents of phlorizin increased and the contents of Ethyl gallate decreased,while there were significant differences between three years old tree and one year old tree leaves.Conclusion The accumulation of polyphenols in leaves of R.chinensis reaches its peak in July to August.The contents of epigallocatechin gallate,epicatechin gallate and phlorizin are higher in young leaves than those in old leaves.The contents of Ethyl gallate are lower and phlorizin are higher in leaves of old trees than those in young trees.
