摘要
为探究褪黑素对高龄小鼠卵母细胞体外成熟的影响,以10~12月龄的ICR雌鼠为试验材料,分别将不同浓度的褪黑素(0、10-4、10-6、10-8 mol·L^(-1))添加至体外成熟培养液中,通过判断卵母细胞的成熟率、受精率及囊胚率筛选出褪黑素最适宜的添加浓度,并通过线粒体分布检测、活性氧水平检测和细胞早期凋亡水平检测,分析褪黑素在卵母细胞成熟中的作用。结果表明:10-6 mol·L^(-1)的褪黑素添加至体外成熟培养液中能有效改善高龄小鼠卵母细胞的成熟、受精和胚胎发育,可通过改善线粒体的分布和活性,降低氧化应激水平和早期细胞凋亡,从而减弱衰老所引起的卵母细胞退化。综合而言,在体外成熟培养液中添加10-6mol·L^(-1)的褪黑素可提高卵母细胞的核质成熟,改善高龄小鼠卵母细胞的质量。
In order to explore the effect of melatonin on in vitro maturation of aged mice oocytes, ICR female mice aged 10-12 months were used as experimental materials. Different concentrations of melatonin(0, 10-4, 10-6, 10-8 mol ·L^(-1)) were added to the in vitro maturation medium. The most suitable concentration of melatonin was selected by oocyte maturation rate, fertilization rate and blastocyst rate. The role of melatonin in oocyte maturation was analyzed by detecting the distribution of mitochondria, the level of reactive oxygen species and the level of early apoptosis. The results showed that the addition of 10-6mol·L^(-1) melatonin significantly increased the maturation rate, fertilization rate and blastocyst rate of aged mice oocytes, and this concentration of melatonin could reduce the oocyte degeneration caused by aging by improving the distribution and activity of mitochondria and reducing the level of oxidative stress and early apoptosis. To sum up, the addition of 10-6mol·L^(-1) melatonin to the in vitro maturation medium can improve the nuclear and cytoplasmic maturation of oocytes and improve the quality of aged mice oocytes.
