摘要
目的:探讨中药复方理心汤对实验性动脉硬化型心肌缺血大鼠炎症因子的影响。方法:将111只雄性SD大鼠随机分成空白对照组13只,模型组23只,西药阳性药组、中药阳性药组以及理心汤高、中、低剂量组各15只,应用腹腔注射维生素D3注射液,高脂饲料喂养及高脂乳剂灌胃方法建立动脉硬化大鼠模型,各药物组均在造模第一天同时开始灌胃给药。8周后,连续3天多点皮下注射异丙肾上腺素,制备大鼠心肌缺血损伤模型,测量白细胞介素(Interleukin,IL)-1、IL-6、基质金属蛋白酶(Matrix Metalloproteinases,MMP)9、肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)、超敏C-反应蛋白(High Sensitivity C-reactive Protein,hs-CRP)因子,并对实验大鼠主动脉、心脏、肝脏进行病理学检查。结果:理心汤高、中剂量组均有降低模型大鼠IL-1含量的趋势,与模型组比较均无明显差异(P>0.05)。理心汤高、中剂量组均能明显降低模型大鼠IL-6含量,与模型组比较均有明显差异(P<0.05)。三个剂量组有降低MMP9含量趋势,但与模型组比较无明显差异(P>0.05)。三个剂量组均能显著降低模型大鼠的hs-CRP、TNF-α含量,与模型组比较均有显著差异(P<0.01或P<0.001)。结论:理心汤对动脉硬化型心肌缺血大鼠炎症因子的影响是治疗冠状动脉粥样硬化性心脏病(简称冠心病)的作用机制之一。
Objective:To explore the effect of the Lixin decoction(理心汤)on inflammatory factors of experimental arteriosclerotic myocardial ischemia rats.Methods:111 male rats were randomly divided into the blank group(13 cases),model group(23 cases),and the Western medicine control group(15 cases),TCM control group(15 cases),high-dose of the Lixin decoction group(15 cases),medium-dose of the Lixin decoction group(15 cases),low-dose of the Lixin decoction group(15 cases).A rat model of arteriosclerosis was established by intraperitoneal injection of vitamin D3 injection,high-fat feed and high-fat emulsion gavage method.In each group,intragastric administration was started simultaneously on the first day of modeling.After 8 weeks,isopropyl adrenaline was given by subcutaneous injection for 3 consecutive days,and myocardial ischemia injury model in rats were prepared.IL-1,IL-6,MMP9,TNF-αand hs-CRP in the rats were measured,and aorta,heart and liver in the experimental rats were examined.Results:In high-dose and medium-dose of the Lixin decoction groups,IL-1 was decreased,and there was no significant difference with the model group(P>0.05).In high-dose and mediumdose of the Lixin decoction groups,IL-1 was decreased,with no statistically significant difference compated with the model group;IL-6 was obvieusly decreased,and there was a significant difference with the model group(P<0.05).In high-dose,medium-dose of and low-dose the Lixin decoction groups,MMP9 was decreased,and there was no significant difference with the model group(P>0.05).In high-dose,medium-dose and low-dose of the Lixin decoction groups,hs-CRP and TNF-αwas decreased,and there was a significant difference with the model group(P<0.01 or P<0.001).Conclusion:The effect of the Lixin decoction on inflammatory factors of atherosclerotic myocardial ischemia rats is one of the mechanisms for the treatment of coronary heart disease.
