雷公藤影响系统性红斑狼疮免疫调节的实验研究 被引量:2

Experimental Study on the Effects of Tripterygium Wilfordii on Immune Regulation of Systemic Lupus Erythema⁃tosus

摘要 目的:观察雷公藤对由Pristane诱发的系统性红斑狼疮(systemic lupus erythematosus, SLE)免疫调节的影响。方法:将40只雌性BALB/c裸鼠按随机原则均分为对照组、SLE组、醋酸泼尼松组(PD组)和雷公藤组(TP组)。除对照组外均采用单次腹腔注射0.5 mL Pristane以诱发SLE模型。3个月后对照组和SLE组给予0.9%氯化钠溶液灌胃(1 mL/鼠),PD组和TP组分别按1次/日、1 mL/鼠剂量给予对应的醋酸泼尼松和雷公藤灌胃治疗4周。末次给药后断尾取静脉血,采用Western Blot检测单核细胞中糖皮质激素受体α(glucocorticoid receptor, GRα)表达,免疫荧光法(IIF)检测静脉血免疫球蛋白G(immunoglobulin G,IgG)、抗史密斯(Sm)抗体、抗双链DNA(dsDNA)抗体、抗核小体抗体(AnuA)、抗增殖性细胞核抗原抗体(PCNA)和抗核糖体P蛋白抗体(ARPA),取脾脏采用免疫荧光法检测B细胞和T细胞亚群CD4~+、CD8~+百分率,取腹腔肉芽肿结节组织并用SP法检测血管通透因子(vascular permeability factor, VPF)和血管内皮生长因子受体1(vascular endothelial growth factor receptor 1,VEGFR1)蛋白、实时定量(RT-PCR)检测VPF和VEGFR1基因表达。结果:与SLE组比,TP组GRα蛋白高表达,抗Sm、dsDNA、AnuA、PCNA和ARPA抗体均减少,IgG、B细胞总数、CD4~+及CD8~+百分率均降低,2组比较差异有统计学意义(P<0.05)。TP组VEGFR1、VPF蛋白及VEGFR1 mRNA/β-actin、VPF mRNA/β-actin灰度值均较SLE组和PD组低(P<0.05),与SLE组和PD组比较差异有统计学意义。结论:雷公藤可能通过调节T、B细胞功能而实现抑制自身免疫性损伤,对由Pristane诱导的SLE具有治疗作用。 Objective To observe the effect of Tripterygium wilfordii on the immune regulation of systemic lupus erythemato⁃sus(SLE)induced by Pristane.Methods Forty female BALB/c nude mice were randomly divided into control group,SLE group,prednisone acetate group(PD group)and tripterygium group(TP group).In addition to the control group,a single intraperitoneal injection of 0.5 ml Pristane was used to induce the SLE model.After 3 months,the control group and the SLE group were given normal saline(1 mL/mouse),and the PD group and the TP group were intragastrically administered with the corresponding prednisone acetate and tripterygic acid at a dose of 1 mL/mouse once a day for 4 weeks.The venous blood was taken from the tail after the last administration.The expression of glucocorticoid receptor(GRα)in monocytes was detected by Western Blot.The immunoglobulin G(IgG),anti-Smith(Sm)antibody,anti-double-stranded DNA(dsDNA)antibody,anti-nucleosome antibody(AnuA),anti-proliferating cell nuclear antigen antibody(PCNA)and anti-ribosomal P protein antibody(ARPA)were detected by immunofluorescence(IIF).The percentages of CD4+and CD8+in B cells and T cell subsets in the spleen were detected by immunofluorescence method.The abdominal granuloma nodules were taken and the vascular permeability factor(VPF)and vascular endothelial growth factor receptor 1(VEGFR1)were detected by SP method,and the gene expression of VPF and VEGFR1 were detected by RT-PCR.Results Compared with the SLE group,the GRαprotein was highly expressed in the TP group,the anti-Sm,dsDNA,AnuA,PCNA and ARPA antibodies were all decreased,and IgG,total number of B cells and the percentates of CD4+and CD8+cells were all reduced.The difference between the two groups was statistically significant(P<0.05).The gray values of VEGFR1,VPF proteins,VEGFR1 mRNA/β-actin and VPF mRNA/β-actin in TP group were lower than those in SLE group and PD group(P<0.05),and there was a significant difference between SLE group and PD group.Conclusion Tripterygium wil⁃fordii can inhibit autoimmune injury by regulating T and B cell function,and has a therapeutic effect on pristane-induced SLE.
