摘要
目的:探究酵母β-葡聚糖加锌复合配方对环磷酰胺诱导免疫抑制幼龄小鼠的免疫调节作用。方法:48只3周龄BALB/c小鼠,随机分成空白组、模型组和复合物组。复合物组灌以由酵母β-葡聚糖和锌组成的复合物(相当于酵母β-葡聚糖:90 mg/kg/d、锌:2.8 mg/kg/d),空白组、模型组灌以等体积蒸馏水,模型组、复合物组在试验的第14、15 d以腹腔注射40 mg/kg环磷酰胺构建免疫低下模型,28 d后,再将小鼠分为两组,第Ⅰ组用于测定免疫器官指数、免疫细胞数目和活性以及细胞因子。第Ⅱ组用于血清溶血素实验和溶血空斑实验。结果:与模型组相比,酵母β-葡聚糖和柠檬酸锌复合配方能显著(P<0.05)提高小鼠免疫器官系数,促使脾淋巴细胞增殖和增加杯状细胞数量,极显著(P<0.01)提高半数溶血值(serum half hemolysis value,HC50)、溶血空斑数、NK细胞活性,增加外周血中α肿瘤坏死因子(Tumor necrosis factor-α,TNF-α)、γ干扰素(interferon-γ,IFN-γ)浓度、白细胞数量,增加派氏结。结论:酵母β-葡聚糖加锌复合配方具有提高体液免疫、NK细胞活性,并增强肠黏膜免疫的作用。
Objective:To study the immunomodulatory effects of yeastβ-glucan and zinc compound formula on cyclophosphamide(CTX)-induced immunosuppression in immature mice.Methods:48 BALB/c mice with 3-week old were randomly divided into three groups:The control group,the model group,and the compound-treated group.The mice in the compound-treated group were treated with yeastβ-glucan and zinc complex(corresponding to yeastβ-glucan:90 mg/kg/d,zinc:2.8 mg/kg/d),while the mice in the control group and model group were fed distilled water in the same volume.On the 14 and 15 d,model group and compound-treated group were injected with CTX to establish immunosuppressive model.After 28 day’s administration,each group was divided into two subgroups.Subgroup I was used to detect immune organ index,numbers and activities of immunocytes and cytokines.Subgroup II was used to conduct serum haemolysin test and hemolytic plaque test.Result:Compared to the model group,yeastβ-glucan and zinc compound formula could significantly(P<0.05)improve mice immune organs indexes,promote the proliferation of spleen lymphocytes and increase the number of intestinal goblet cell,also could highly significantly(P<0.01)improve the serum half hemolysis value(HC50),increase the number of hemolytic plaques,enhance NK cell activity,raise the concentration of tumor necrosis factor-α(TNF-α)and interferon-γ(IFN-γ)in peripheral blood,the numbers of white blood cell(WBC),Peyer's patches.Conclusion:The yeastβ-glucan and zinc compound formula had the function of enhancing humoral immunity,NK cell activity and intestinal mucosa immunity in immature mice.
