基于网络药理学探讨三物备急方保留灌肠护理肠梗阻的作用机制 被引量:4

Mechanism of action of Sanwu Beiji prescription in retention enema for intestinal obstruction: A study based on network pharmacology

摘要 目的:探讨三物备急方保留灌肠护理肠梗阻的作用机制。方法:借助中药系统药理学分析平台(TCMSP)等多个数据库联合检索三物备急方中化学成分和作用靶点,通过UniProt、HCBI和PubMed等数据库查询靶点对应的基因,进而运用Cytoscape软件构建化合物-靶点网络筛选出核心靶点,最后通过基因本体(GO)功能富集分析研究其作用机制。结果:共筛选出了9个活性化合物,其中化合物-靶点(基因)网络包含9个活性化合物和相应靶点46个,核心靶点涉及前列腺素内过氧化物合酶1(PTGS1)等。结论:三物备急方保留灌肠系通过多靶点、多通路来治疗肠梗阻,这为进一步深入探讨其作用的物质基础以及作用机制提供了预测作用,并为临床使用三物备急方保留灌肠护理肠梗阻患者提供了依据。 Objective: To investigate the mechanism of action of Sanwu Beiji prescription in retention enema for intestinal obstruction.Methods: Several databases including Traditional Chinese Medicine Systems Pharmacology were searched for the chemical components of Sanwu Beiji prescription and their targets, and the databases including UniProt, HCBI,and PubMed were searched to obtain the corresponding genes of these targets.Cytoscape software was used to construct a component-target network and screen out core targets, and finally a gene ontology functional enrichment analysis was used to investigate the mechanism of action.Results: A total of 9 active components were screened out.The component-target(gene) network contained 9 active components and 46 corresponding targets, and the core targets involved prostaglandin-endoperoxide synthase 1.Conclusion: Retention enema with Sanwu Beiji prescription exerts a therapeutic effect on intestinal obstruction via multiple targets and pathways, which helps to further explore its material basis and mechanism of action and provides a basis for the application of retention enema with Sanwu Beiji prescription in patients with intestinal obstruction.
