摘要
目的探究LY2109761对大鼠肝星状细胞(HSC-T6)生存与凋亡的影响。方法体外培养至对数生长期的HSC-T6细胞分为空白组、LY2109761给药组和阳性组(25μmol/L秋水仙碱),采用CCK-8法检测细胞存活率,细胞划痕法观察细胞迁移能力,流式细胞术检测细胞凋亡率,Western blot法检测α-SMA、Collagen Ⅰ、TGF-β1和p-Smad2的蛋白表达水平。结果与空白组比较,LY2109761给药组和阳性组细胞的存活率和细胞迁移能力降低(P <0.05或P <0.01),细胞凋亡率明显增加,差异有统计学意义(P <0.01),且LY2109761给药组和阳性组细胞的α-SMA、Collagen I、TGF-β1和p-Smad2蛋白表达水平均降低,差异有统计学意义(P <0.05或P <0.01)。结论 LY2109761可能通过抑制TGF-β1/Smad通路、增加HSC-T6细胞凋亡率,抑制HSC-T6细胞的生存及活化,从而产生抗纤维化作用。
Objective To explore the effect of LY2109761 on the survival and apoptosis of Hepatic Stellate Cell-T6( HSC-T6). Methods Culturing HSC-T6 cells in vitro to logarithmic growth phase and dividing the cells into blank group,LY2109761-treated group and positive group( 25 μmol/L colchicine). Cell viability was detected by CCK8 method;cell migration ability was tested by Cell Scratch Assay;apoptosis was assessed by flow cytometry;protein expression levels of α-SMA,Collagen Ⅰ,TGF-β1,and p-Smad2 in HSC-T6 cells were detected by Western blot. Results Compared with the blank group,the cell viability rate and cell migration ability of the LY2109761-treated group and positive group were significantly reduced and the difference was statistically significant( P < 0. 05 or P < 0. 01);the apoptosis rate was increased markedly( P < 0. 01),while the expression levels of α-SMA,Collagen I,TGF-β1,and p-Smad2 of HSC-T6 cells in the LY2109761-treated group and positive group were significantly decreased and the difference was statistically significant( P < 0. 05 or P < 0. 01). Conclusion By inhibiting the TGF-β1/Smad pathway,LY2109761 may increase the apoptosis rate of HSC-T6 cells,inhibit the survival and activation of HSC-T6 cells,and thus producing an anti-fibrosis effect.
