石刁柏愈伤组织对茎枯病菌毒素胁迫的生理响应

摘要 为阐明石刁柏愈伤组织在茎枯病菌毒素胁迫下的生理生化响应,本研究利用茎枯病菌毒素胁迫石刁柏‘冠军’的愈伤组织,研究了石刁柏愈伤组织的过氧化氢(H2O2)积累、组织细胞结构、DNA条带、植物激素及蛋白质变化。结果表明,在茎枯病菌毒素胁迫下,石刁柏愈伤组织出现H2O2积累、细胞空泡化和DNA片段化等现象,表现出细胞过敏反应(hypersensitive response,HR)和细胞程序性死亡(programmed cell death PCD)的特征。植物激素生长素(IAA)、脱落酸(ABA)和玉米素核苷(ZR)含量升高,赤霉酸(GA3)含量降低;可溶性蛋白含量呈先显著升高,后期小幅缓慢下降趋势;蛋白质电泳分析显示,在胁迫48 h时出现了2条新的特征条带,这两条特征条带随着胁迫时间的延长逐渐加深,表达量增加。表明石刁柏愈伤组织与茎枯病菌毒素互作过程中,发生了HR和PCD,产生了病程相关蛋白;较高水平的IAA、ZR和ABA和较低水平的GA3参与了石刁柏愈伤组织的抗病响应过程。本研究揭示了石刁柏愈伤组织在茎枯病菌毒素胁迫下的生理生化响应,为石刁柏抗茎枯病突变体的离体筛选提供了参考和依据。 In order to reveal the physiological and biochemical response of the callus of Asparagus officinalis L.to the toxin of stem blight pathogen, the accumulation of hydrogen peroxide(H2O2), histiocytic structure, DNA bands, endogenous hormones and proteins in asparagus callus were studied under the stress of the toxin of stem blight pathogen. The results showed that H2O2 accumulation, cell vacuolization and DNA fragmentation appeared in the callus of asparagus under the stress of toxin, which showed the characteristics of hypersensitive response(HR) and programmed cell death(PCD). The contents of indole acetic acid(IAA), abscisic acid(ABA) and zetin-riboside(ZR) increased, while the gibberellin A3(GA3) decreased. The soluble protein increased significantly at first and decreased slowly at later stage. Protein electrophoresis analysis showed that 2 new characteristic bands appeared at 48 hours of stress, and their expression increased with the stress time. The results indicated that HR and PCD occurred and pathogenesis-related proteins were produced in asparagus callus during the interaction with the toxin of stem blight pathogen, and higher levels of IAA, ZR, ABA and lower levels GA3 were involved in the response. This study revealed the physiological and biochemical response of the asparagus callus to the toxin of stem blight pathogen, and provided a reference and basis for the in vitro screening of asparagus callus mutants resistant to stem blight.
