摘要
目的建立基于江香薷有效基准特征图谱的质量表征模式、方法,以精准、有效地评价江香薷饮片质量。方法以确定有活血药效的江香薷饮片(饮片编号S5)作为有效饮片基准,以有效特征代表性成分及其含量作为质量表征基准点,采用HPLC-PDA建立其有效基准特征图谱,以对照品模拟基准点的有效基准特征图谱质量表征信息作为评价江香薷饮片有效质量基准标尺,并对其准确性进行佐证及质量评价应用与验证。结果以江香薷饮片S5(饮片基准)中有效特征代表性成分黄芩素-7-甲醚作为质量表征基准点,并以其对照品对基准点进行准确模拟(保留时间:61.020分钟、检测溶液含量:0.0115 mg/mL),分别对江香薷有效饮片S5(基准)进行质量表征,结果佐证了检测方法的可行性及检测结果的准确性。以模拟基准点为基准得到江香薷有效基准特征图谱质量表征信息:15个特征峰的相对保留时间、相对峰面积及7个特征代表性成分(咖啡酸、异迷迭香酸苷、野黄芩苷、迷迭香酸、黄芩素-7-甲醚、香荆芥酚、麝香草酚)含量、相对含量、含量换算系数等,作为评价江香薷饮片的质量基准标尺。其15个特征峰及其7个特征代表性成分涵盖了江香薷药学架构酚酸类、黄酮类、萜类三种主要化学类型,14批江香薷饮片的特征图谱中均含有15个共有特征峰,且基于含量换算系数得到的14批江香薷饮片中7个特征代表性成分的含量与实测含量基本一致;采用所构建的江香薷质量评价模式及其有效质量基准标尺,对14批江香薷饮片的质量进行评价得出:编号S13、S3、S8、S2、S1、S10、S12、S5(饮片基准)优良度居前。结论建立的基于江香薷有效基准特征图谱质量表征模式与评价方法,可简易、准确、整体地对江香薷饮片进行质量表征与评价,为江香薷饮片的精准质量评价与有效应用提供科学依据,亦为其他中药的质量表征与评价提供借鉴。
Objective To establish the quality characterization mode and method based on the effective reference characteristic spectrum of Moda chinensis Maxim.cv.jiangxiangru,so as to accurately and effectively evaluate the quality of Moda chinensis Maxim.cv.jiangxiangru.Methods To determine prepared Moda chinensis Maxim.cv.jiangxiangru pieces with efficacy of blood-invigorating as effective decoction pieces(benchmark),effective characteristics of typical components and their content as quality characterization of datum,the HPLC-PDA to establish the effective benchmark characteristic spectrum,are made valid by means of the reference substance simulation benchmark of datum characteristic spectrum quality characterization information as evaluation benchmark Moda chinensis Maxim.cv.jiangxiangru decoction pieces effective mass scale,and to support the accuracy and quality evaluation of application and validation.Results With Moda chinensis Maxim.cv.jiangxiangru decoction pieces S5(decoction pieces benchmark)effective characteristics of typical components in radix scutellariae element-7-methyl ether as quality characterization of datum,and with the reference substance to the reference point for accurate simulation(retention time:61.020 min,to detect the contents of solution:0.0115 mg/mL),respectively,for Moda chinensis Maxim.cv.jiangxiangru effective decoction pieces(benchmark)for quality characterization,the results support the feasibility of detection method and the accuracy of the test results.To simulate a benchmark as a benchmark for Moda chinensis Maxim.cv.jiangxiangru effective datum charateristic spectrum quality characterization information:15 relative retention time and relative peak area of characteristic peak and the seven characteristics of typical components(caffeic acid,salviaflaside,scutellarin,rosmarinic acid,negletein,carvacrol,thymol)content,relative content,content conversion factor,etc.,as the assessment of the quality of Moda chinensis Maxim.cv.jiangxiangru decoction pieces benchmark scale.Its 15 characteristic peak and seven characteristics of typical components covers Moda chinensis‘Jiangxiangru’medicine architecture phenolic acids,flavonoids,terpenoids,three main chemical types,14 batches of characteristic spectrum of Moda chinensis Maxim.cv.jiangxiangru decoction pieces contains 15 common characteristic peak,and based on the content of the conversion coefficient of 14 batches of Moda chinensis Maxim.cv.jiangxiangru decoction pieces content of seven representative composition characteristics in basic agreement with the measured content;Based on the quality evaluation model of Moda chinensis Maxim.cv.jiangxiangru decoction pieces and its effective quality reference scale,the quality evaluation of 14 batches of Moda chinensis Maxim.cv.jiangxiangru decoction pieces was carried out,and it was concluded that the quality of No.S13,S3,S8,S2,S1,S10,S12 and S5(decoction pieces benchmark)was in the first place.Conclusion The quality characterization model and evaluation method based on the effective benchmark characteristic spectrum of Moda chinensis Maxim.cv.jiangxiangru can easily,accurately and comprehensively conduct the quality characterization and evaluation of Moda chinensis Maxim.cv.jiangxiangru,which provides scientific basis for the precise quality evaluation and effective application of Moda chinensis Maxim.cv.jiangxiangru,and also provides reference for the quality characterization and evaluation of other Traditional Chinese medicine.
