益生木质纤维素对鸡盲肠黏膜免疫屏障的影响 被引量:4

摘要 试验选用9周龄体质量相近的健康边鸡和伊莎褐鸡各108只笼养,采用2因素3水平试验设计,育成期自行配制益生木质纤维素含量不同、等能等蛋白的饲料,益生木质纤维素设2个水平(2%和4%),研究益生木质纤维素对不同品种鸡盲肠黏膜免疫屏障的影响。结果表明,20周边鸡胸腺免疫器官指数极显著高于伊莎褐鸡,且边鸡促炎性细胞因子TNF-α、IL-6 mRNA表达水平有低于伊莎褐鸡的趋势。添加益生木质纤维素对20周盲肠绒毛高度和绒隐比无显著影响,但对隐窝深度、黏液层厚度、IL-1、TNF-α、IL-10、IL-6、MUC2 mRNA表达有显著影响,且2%益生木质纤维素处理组极显著增加了盲肠黏液层厚度以及IL-10、MUC2 mRNA表达,极显著降低了盲肠隐窝深度和TNF-αmRNA表达,显著降低了盲肠IL-1、IL-6 mRNA表达;品种与益生木质纤维素互作对20周盲肠黏液层厚度以及TNF-α、MUC2 mRNA表达有显著影响。添加低纤维组(2%益生木质纤维素)可改善伊莎褐鸡和边鸡盲肠黏膜免疫屏障。 To investigate the effects of probiotic lignocellulosic(OptiCell)on the mucosal immune barrier of cecum in chickens of different breeds 108 healthy local Bian Chickens and ISA Brown Chickens of the same age and body quality were selected to be raised in a cage.Using a two factors with three levels experimental design and each chicken was randomly divided into 3 treatments,each treatment was 6 replicates,and each repeat was 6 chickens.During the incubation period(9-20 weeks),a self-prepared feed with isoprotein was prepared.Treatment 1(OL)and 2(OH)contained 2%and 4%OptiCell.The adjustment period lasted for 1 weeks,and the experimental period lasted for 11 weeks.The results showed that at 20 weeks,the index of thymus immune organs in Bian Chickens was significantly higher than that ISA Brown Chicken(P<0.01),and the expression levels of TNF-αand IL-6 mRNA in Bian Chickens were lower than that ISA Brown Chicken.Add OptiCell had no significant effect on villus height and villi height/crypt depth at 20 weeks,but had a significant effect on crypt depth,mucus layer thickness,TNF-α,IL-1,IL-10,IL-6,MUC2 mRNA expression,and OL group significantly increased the mucus layer thickness and IL-10,MUC2 mRNA expression(P<0.01),significantly reduced the crypt depth and TNF-αmRNA expression(P<0.01),significantly reduced the cecum IL-6 and IL-1 mRNA expression(P<0.05).At 20 weeks,the interaction between the OptiCell and the variety had significant effects on the thickness of the mucus layer and the expression of the pro-inflammatory cytokines TNF-a and MUC2 mRNA(P<0.05).In summary,the addition of low-fiber group 2%OptiCell can improve the mucosal immune barrier of the cecum of ISA Brown Chicken and Bian Chicken.
