补阴牵正方对帕金森病细胞模型氧化应激损伤的保护作用及机制研究 被引量:4

Study on protective effect and mechanism of Buyin Qianzheng Formula on oxidative stress injury in Parkinson’s disease cell model

摘要 目的:通过对补阴牵正方抗氧化应激减少帕金森病(PD)细胞模型凋亡的研究,探讨中药治疗PD的作用机制。方法:利用1-甲基-4-苯基吡啶离子(MPP+)诱导构建PD细胞模型,将SH-SY5Y细胞分为正常组、模型组和中药组。采用CCK-8法检测细胞存活率,流式细胞仪检测细胞凋亡率和活性氧(ROS)水平,酶标仪检测细胞丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力,Western blot检测细胞色素C(Cyt C)蛋白的表达。结果:与正常组比较,MPP+处理后,细胞存活率和SOD活力显著下降(P<0.01),细胞凋亡率、ROS水平、MDA含量及Cyt C蛋白表达水平均显著增高(P<0.01)。与模型组比较,中药干预后,PD细胞模型存活率和SOD活力显著上升(P<0.01),凋亡率、ROS水平、MDA含量及Cyt C蛋白表达水平均显著降低(P<0.01)。结论:补阴牵正方可以改善氧化应激,减少PD细胞模型凋亡,从而发挥对PD的神经保护作用。 Objective:Study on decreasing apoptosis of PD cell model by anti-oxidative stress of Buyin Qianzheng Formula,to explore the mechanism of Chinese medicine in treating PD.Methods:The PD cell model was constructed by MPP+induction,and SH-SY5 Y cells were divided into normal group,model group,and Chinese medicine group.Cell survival rate was determined by CCK-8 method,cell apoptosis rate and ROS level were detected by flow cytometry,MDA content and SOD activity were detected by enzyme marker,and Cyt C protein expression was detected by Western blot.Results:Compared with the normal group,after MPP+treatment,cell survival rate and SOD activity was notably decreased(P<0.01),apoptosis rate,ROS level,MDA content and Cyt C protein expression levels were prominently increased(P<0.01).Compared with the model group,after the intervention of Buyin Qianzheng Formula,the survival rate of PD cell model and the activity of SOD was obviously increased(P<0.01),the apoptosis rate,levels of ROS,MDA and Cyt C protein expression levels were significantly decreased(P<0.01).Conclusion:Buyin Qianzheng Formula can improve the neuroprotective effect of PD by improving oxidative stress and reducing apoptosis of PD cell model.
