头针对中风后肢体痉挛大鼠脑皮质中环磷腺苷效应元件结合蛋白影响的研究 被引量:4

摘要 目的探讨环磷腺苷效应元件结合蛋白(CREB)在中风后肢体痉挛大鼠脑皮质神经元细胞恢复中的作用机制,以及头针对中风后肢体痉挛的治疗效果。方法将45只Sprague Dawley大鼠(SD大鼠)随机分成假手术组15只,实验组30只。实验组采用线栓法进行大脑中动脉闭阻模型(Middle Cerebral Artery Occlusion,MCAO)造模。将造模成功大鼠随机分到模型组和头针组,给予相应的治疗1周,并每日观察记录改良Ashworth评分、平衡木行走实验评分等动物行为学数据。1周后对大鼠脑组织进行取材,采用尼氏染色观察缺血损伤病变部位的神经元细胞及尼氏小体的变化,并采用Western Blot、PCR检测脑皮质中的CREB、p-CREB及其mRNA的表达。结果行为学检测结果显示,与假手术组比较,模型组大鼠改良Ashworth评分增高,平衡木行走实验评分降低;与模型组比较,头针组大鼠改良Ashworth评分降低,平衡木行走实验评分增高。头针组大鼠脑皮质神经元排列整齐且尼氏体数量增加,CREB及p-CREB蛋白表达较模型组增加。头针组大鼠脑皮质CREB mRNA表达较模型组增加。结论头针调控对脑皮质内CREB表达水平,促进缺血损伤组织的神经元细胞恢复,改善肢体痉挛状态,促进运动功能恢复。 Objective To investigate the mechanism of cyclic AMP response element binding protein(CREB)in the recovery of cerebral cortex neurons in rats with limb spasticity after stroke,and the therapeutic effect of scalp acupuncture on it.Methods 45 Sprague Dawley rats(SD rats)were randomly divided into sham operation group(n=15)and experimental group(n=30).In the experimental group,the middle cerebral artery occlusion(MCAO)model was made with the line occlusion method.The successfully modeled rats were randomly divided into model group and scalp acupuncture group,and were given corresponding treatment for 1 week.Animal behavior data such as modified Ashworth score and balance beam walking test score were observed and recorded daily.One week later,the rats brain tissues were collected,and the changes of neuron cells and Nissella bodies in the ischemic lesion site were observed by Nissl’s staining,and the expressions of CREB,p-CREB and their mRNA in the cerebral cortex were detected by Western Blot and PCR.Results The behavioral test results showed that compared with the sham operation group,the modified Ashworth score of the model group increased,while the balance beam walking test score decreased.Compared with the model group,the modified Ashworth score of the scalp acupuncture group decreased,while the balance beam walking test score increased.The cerebral cortex neurons in the rats of the scalp acupuncture group were neatly arranged and the number of Nissella bodies was increased,and the protein expressions of P-CREB and BDNF were increased compared with those in the model group.The cerebral cortex CREB and mRNA expression in the rats of the scalp acupuncture group was increased compared with that of the model group.Conclusion Scalp acupuncture can regulate the CREB expression level in cerebral cortex,promote the recovery of neurons in ischemic damaged tissues,improve limb spasm,and promote the recovery of motor function.
