摘要
【目的】了解成年牦牛(Bos grunniens)菱脑中促红细胞生成素(erythropoietin,EPO)及其受体(erythropoietin receptor,EPOR)的表达分布特征,对其与低氧适应性之间的关系进行探讨.【方法】利用qRT-PCR及免疫组织化学技术,对EPO及EPOR在牦牛菱脑中的表达与分布进行研究.【结果】EPO和EPOR mRNA在成年牦牛小脑蚓部的相对表达量最高,EPO及EPOR蛋白主要表达于小脑皮质浦肯野氏细胞胞质.【结论】在低氧适应过程中,EPO及EPOR在牦牛菱脑蚓前叶的表达量最高,因此牦牛在低氧状况下其小脑蚓前叶可能发挥更重要的功能作用.
【Objective】The study aims to explore on the expression and distribution of erythropoietin(EPO)and its receptor(erythropoietin receptor,EPOR)in adult yak(Bos grunniens)rhombencephalon.The research also discuss the relationship between EPO and EPOR and the adaptation to hypoxia.【Method】Qualitative real time polymerasre chain reaction(qRT-PCR)and immunohistochemical(IHC)methods were employed to examine the distribution and expression of EPO and EPOR in the Rhombencephalon of the adult yak.【Result】The relative expression of EPO and EPOR mRNA in adult yak cerebellar vermis showed the highest while the EPO and EPOR proteins main expression was found in the cytoplasm of Purkinje cells in cerebellar cortex.【Conclusion】In the process of hypoxia adaptation,EPO and EPOR have the highest expression levels in the archicerebellum of yak rhombus.Therefore,the archicerebellum in yak might play an important functional role under hypoxic conditions,and its specific regulatory mechanism needs to be further studied.
