双刺参总皂苷的纯化及其体外活性 被引量:1

Purification of Total Saponins of Shuangcishen and Its Activity in Vitro

摘要 以吸附率及解吸率为考察指标,探索双刺参总皂苷的纯化工艺,并对双刺参总皂苷的体外抗肿瘤活性进行研究。通过静态吸附-解吸试验、静态吸附及解吸动力学试验筛选适合纯化双刺参总皂苷的大孔树脂类型;采用单因素试验及正交试验,对D-101型大孔树脂纯化双刺参总皂苷的工艺条件进行优化;通过对亚硝酸盐的清除及阻断亚硝胺的合成活性,对其体外抗肿瘤活性进行评价。结果表明,双刺参总皂苷的最优纯化工艺为:D-101型大孔树脂中加入7倍量,总皂苷质量浓度1 mg/mL供试液,室温振荡吸附4 h,以22倍量的75%乙醇振荡解吸30 min,经工艺纯化,双刺参提取物中总皂苷的含量由17.83%提高到37.04%;双刺参总皂苷对清除亚硝酸盐和阻断亚硝胺合成具有一定活性,说明其具有体外抗肿瘤作用。试验结果为双刺参胶囊的研究与开发提供基础数据。 Based on the index of adsorption and desorption, the purification process of total saponins of Shuangcishen(TSS) was explored, and its antitumor activity in vitro was studied. The type of macroporous resin which was suitable for the purification of TSS was screened by notonlystatic adsorption-desorption test but also static adsorption and desorption kinetics test. The single-factor experiment and orthogonal experiment were used to optimize the process conditions for purification of TSS with D-101 macroporous resin. The antitumor activity in vitro was evaluated by eliminating nitrite and blocking the synthesis of nitrosamine. The optimal purification process of TSS was obtained. D-101 macroporous resin was added with a 7-fold amount of sample solution with total saponin mass concentration of 1 mg/mL, which was absorbed by shaking at room temperature for 4 h and desorbed by 22-fold amount of 75% ethanol for 30 min. After purification by the above process, the content of total saponins in the extract of Shuangcishen increased from 17.83% to 37.04%. The TSS had certain activity of removing nitrite and blocking nitrosamine synthesis which indicated that it had antitumor activity in vitro. The results provided basic data for further research and development of Shuangcishen capsule.
