摘要
F-box基因在植物组织器官发育及非生物胁迫响应中起着重要的作用。为进一步探讨其作用机制,从结球甘蓝品种BoJF-16-1中利用反转录PCR(RT-PCR)技术克隆得到一个F-box基因BoFBX117,其cDNA全长为936 bp,编码311个氨基酸,蛋白分子量为30.14 kDa,等电点为9.71。该基因编码BoFBP7蛋白,序列同源性分析表明,BoFBP7与芜菁、萝卜、拟南芥的FBP7蛋白亲缘关系较近,同源性分别为99.36%、98.39%、92.26%;实时荧光定量PCR(RT-qPCR)分析表明,BoFBX117基因在结球甘蓝莲座叶中最高,其次是根,幼叶中表达量最低,表现出组织特异性;在低温胁迫处理下,该基因表达量总体呈现上升趋势,说明该基因的表达受低温胁迫诱导。推测BoFBX117基因可能在结球甘蓝叶片发育及低温胁迫中发挥重要的生物学功能。本研究为深入解析F-box基因调控植物生长发育及非生物逆境胁迫的分子机制提供了依据。
F-boxgene plays an important role in plant tissue development and abiotic stress response.To further explorethe mechanism,a F-box geneBoFBX117 was cloned from the cabbage variety"BoJF-16-1"by RT-PCR.Its cDNAwas 936 bp in length,encoding 311 amino acids,protein molecular weight was 30.14 kDa,and isoelectric point was9.71.The gene encodes BoFBP7 protein.Sequence homology analysis showed that BoFBP7 is closely related to otherFBP7 proteins in turnip,radish and Arabidopsis,with homology of 99.36%,98.39%,and 92.26%,respectively.Quantitative PCR analysis showed that the expression ofBoFBX117 gene was the highest in the rosette leaves,followedby the roots,and the lowest was in the young leaves,suggesting the tissue specific expression ofBoFBX117 gene incabbage.Under low temperature stress,the expression ofBoFBX117 gene showed an upward trend,indicating that theexpression of this gene was induced by low temperature stress.It is suggested that theBoFBX117 gene may play animportant role in leaf development and response to low temperature stress in cabbage.This will provide basis for furtheranalysis of the molecular mechanism of F-box gene regulating plant development and abiotic stress.
