摘要
目的基于网络药理学探讨白果治疗哮喘作用机制。方法基于TCMSP数据库获取白果化学成分及其对应靶点,基于OMIM、Gene Cards数据库获取哮喘相关靶点,利用Venny2.1绘制白果靶点与哮喘疾病靶点的韦恩图,利用Cytoscape3.6.1软件构建中药-靶点网络及应用STRING平台构建蛋白相互作用网络(PPI),最后,应用R语言软件对关键靶点进行GO富集分析和KEGG通路富集分析。结果共获得14个化合物,199个药物与哮喘相关靶点,有效成分中度值较高的为紫草酮(Shikodonin),奎尼辛(Quinicine)及芒柄花黄素(formononetin)等。PPI网络中核心基因为AKT丝氨酸/苏氨酸激酶(AKT1)、白细胞介素-6(IL-6)、丝裂原活化蛋白激酶3(MAPK3)等;GO富集分析显示DNA结合转录激活活性,RNA聚合酶Ⅱ特异性(DNA-bindingtranscription activator activity,RNA polymeraseⅡ-specific)为显著性最高的通路,其次是受体配体活性(receptor ligand activity)等;KEGG富集通路分析显示人巨细胞病毒感染为显著性最高的通路,其次是PI3K-Akt信号通路等。结论该研究系统地探讨了白果治疗哮喘的作用机制,为今后进一步实验验证提供了依据。
Objective Based on the network pharmacology,to explore the mechanism of Baiguo(Ginkgo Semen) in the treatment of asthma.Methods The chemical components and corresponding targets of Baiguo(Ginkgo Semen) were obtained based on TCMSP database and asthma related targets were obtained based on OMIM and gene cards database.Venny2.1 was used to draw the Wayne map of Baiguo(Ginkgo Semen) targets and asthma disease targets.The Chinese medicine target network was constructed by using Cytoscape 3.6.1 software and the protein-protein interaction network(PPI) was constructed by using string platform.Finally,R language software was applied to key points.GO enrichment analysis and KEGG pathway enrichment analysis were carried out for the target.Results A total of 14 compounds and 199 asthma related targets were obtained.Shikodonin,quinicine and formononetin were the most effective components.The core genes of PPI network were Akt serine/threonine kinase(AKT1),interleukin-6(IL-6),mitogen activated protein kinase 3(mapk3),etc.GO enrichment analysis showed DNA binding transcription activation activity and RNA polymerase Ⅱ-specific was the most significant pathway,followed by receptor ligand activity.KEGG enrichment pathway analysis showed that human cytomegalovirus infection was the most significant pathway,followed by PI3 K Akt signaling pathway.Conclusion This study systematically explored the mechanism of Baiguo(Ginkgo Semen) in the treatment of asthma,and provided the basis for further experimental verification in the future.
