摘要
为探究黑枣遗传多样性及表型变异规律,对“冀洪1号”黑枣与普通黑枣的叶片形态差异进行了研究。叶片扫描成像后,使用Lamina软件对4个叶片形态指标进行测量(叶面积、叶周长、叶长和叶宽),计算其统计参数;使用Image J软件对叶片形态进行标点并通过Morpho J软件进行主成分分析(PCA)、薄板样条分析及判别分析。方差分析结果显示,“冀洪1号”黑枣与普通黑枣的叶形性状存在较大的种间差异,“冀洪1号”黑枣的叶片较大,但其变异幅度较小。基于Morpho J软件叶形变异主成分分析的结果显示,第一主成分(PC1)和第二主成分(PC2)共占总体变量的63.447%(分别为50.537%和12.910%),薄板样条网格图结果显示,“冀洪1号”黑枣与普通黑枣形态相似,仅在叶柄及叶片最大宽度处有微小差异。基于Lamina软件和Morpho J软件对“冀洪1号”黑枣与普通黑枣叶片进行比较,能够找出叶片间的微小差别,并且对发现叶形变异与植物遗传多样性的关系有积极作用。
This study intends to reveal the leaf morphological variation of"Jihong No.1"and ordinary Diospyros lotus L.,and provide a basis for exploring their genetic relationship.After leaf scan imaging,the Lamina software was used to measure 4 leaf morphological indi-cators(leaf area,leaf circumference,leaf length and leaf width)and calculate the statistical parameters.Image J software was used to punctuate the leaf morphology and Morpho J software was used to perform principal component analysis(PCA),thin plate spline analysis and discriminant analysis.The results of variance analysis showed that the leaf shape traits of"Jihong No.1"and ordinary Diospyros lotus L.had large differences between species.The leaves of"Jihong No.1"were larger,but the variation range was small.Based on the results of the principal component analysis of leaf shape variation by Morpho J software,the first principal component(PC1)and the second principal component(PC2)together accounted for 63.447%of the overall variables(50.537%and 12.910%respectively).The results of the thin-plate spline grid map showed that"Jihong No.1"was similar in morphology to ordinary Diospyros lotus L.,with only minor differences in the petiole and maximum leaf width.Based on Lamina software and Morpho J software,the comparison of the leaves of"Jihong No.1"and ordinary Diospyros lotus L.shows the small differences between the leaves,and has a positive effect on discovering the relationship between leaf shape variation and plant genetic evolution.
