拜颤停复方在大鼠脑组织中的药代动力学研究 被引量:2

摘要 目的:利用UPLC-MS研究拜颤停复方中代表性成分异嗪皮啶、紫丁香苷、刺五加苷E、芍药苷与异钩藤碱在大鼠脑组织中的动态变化过程。方法:SD大鼠72只,随机分为正常组与模型组,模型组腹腔注射MPTP连续7 d造模,正常组给予等量生理盐水,分别于第8天给予拜颤停复方,并于给药前与给药后的0.16、0.5、1、2、4、6、8、10、12、24和48 h处死大鼠并取出脑组织,处理后进样分析。结果:各目标成分在帕金森(PD)模型脑组织中的浓度均大于生理状态大鼠脑组织中的浓度,且与空白组相比,模型组各代表性成分的Cmax与AUC均有显著升高。结论:拜颤停复方在大鼠体内吸收迅速,能够快速分布于脑组织,且在病理模型中药物的选择性作用更为明显,结果可为拜颤停复方新药开发及临床应用等方面提供实验依据。 Objective:A UPLC-MS method was developed for the simultaneous determination of isoprazine,syringin,acanthopanax E,paeoniflorin and isorhynchophylline,which were the representative components in Baichanting Compound,in rat brain tissue.Methods:72 SD rats were randomly divided into the normal group and the model group.The model group was injected intraperitoneally with MPTP for 7 consecutive days,whereas the normal group was given the same amount of normal saline,and Baichanting Compound was administrated on the 8th day.The brain tissue was collected at 0.16 h,0.5 h,1 h,2 h,4 h,6 h,8 h,10 h,12 h,24 h and 48 h after the administration for the analysis.Results:The concentrations of each target component in the brain tissue of the PD model were greater than those in the physiological state of the rat brain tissue,and the Cmax and AUC of the representative components of the model group were significantly increased compared to those in the blank group.Conclusion:Baichanting Compound is rapidly absorbed in rats,and can be quickly distributed in brain tissue.The selective effect of the drug is more obvious in pathological models.The results can provide experimental basis for the development of new medicine and clinical application of Baichanting Compound.
