摘要
目的优选银桑颗粒的提取工艺、挥发油包合工艺。方法以出膏率及总黄酮提取量为考察指标,采用水蒸气蒸馏法,单因素考察料液比、提取时间对银桑颗粒的提取影响;以出膏率及总黄酮、绿原酸提取量为综合评价指标,采用L_(9)(3^(4))正交试验考察提取时间、料液比、提取次数对水煎煮工艺的影响;以薄荷油包合物收得率、包合率为综合评价指标,采用L_(9)(3^(4))正交试验优化薄荷油与β-环糊精的比例、β-环糊精与水的比例、包合温度、包合时间对挥发油包合工艺的影响。结果水煎煮最佳提取工艺为加12倍量水,煎煮3次,每次1 h;包合最佳工艺为薄荷油与β-环糊精的比例是1∶10,β-环糊精与水的比例是1∶10,包合温度为45℃,包合时间为3 h。结论优选的提取工艺提取率高、重复性好,挥发油包合工艺稳定可行,适用于银桑颗粒的工业化生产。
Objective To optimize the extraction process and volatile oil inclusion process of Yinsang Granules(银桑颗粒).Methods The extract rate and total flavonoids were used as the indexes,and the effects of the ratio of material to liquid and the extraction time on the extraction of Yinsang Granules were investigated by steam distillation.The effects of extraction time,solid-liquid ratio and extraction times on the decoction process were investigated by L_(9)(3^(4))orthogonal test.The effects of the ratio of menthol oil toβ-cyclodextrin,the ratio ofβ-cyclodextrin to water,the inclusion temperature and the inclusion time on the inclusion process of volatile oil were optimized by L_(9)(3^(4))orthogonal test.Results The optimal extraction process was to add 12 times the amount of water and decoction for 3 times,1 h for each time.The optimal inclusion technology of volatile oil included the ratio of volatile oil andβ-cyclodextrin was 1∶10.The ratio ofβ-cyclodextrin and water was 1∶10.The inclusion temperature was 45℃and the inclusion time was 3 h.Conclusion The optimized extraction process has high extraction rate and good repeatability.The optimal extraction technology of volatile oil and inclusion technology are stable and feasible,and are suitable for the industrial production of Yinsang Granules.
