摘要
为探究菟丝子水提物对子宫内双酚A(bisphenol A,BPA)暴露小鼠断乳时期的生殖保护作用,将50只妊娠母鼠随机分为5组,每组10只,在孕期制造小鼠子宫内BPA暴露模型,并给予不同剂量的菟丝子治疗,直到孕鼠生产,记录母鼠孕期(d)和孕期增重(g);子代小鼠出生后,记录仔鼠总数(只)、出生窝重(g)、出生均重(g)及死胎数(只);小鼠21日龄(断乳)时,称重后眼球采血,脱颈处死,统计仔鼠雌雄比、卵巢系数;ELISA检测雌仔鼠血清中雌二醇(E2)、孕酮(P4)、BPA含量及卵巢雌激素受体α(ERα)水平;实时定量荧光PCR法(RT-qPCR)对卵巢DNA甲基转移酶3a(Dnmt3a)、DNA甲基转移酶3b(Dnmt3b)、ERα和孕酮受体(PR)mRNA相对转录水平进行检测。结果显示,BPA饮水可显著提高子代小鼠的死胎率及血清中BPA水平(P<0.05),显著降低子代雌鼠血清E2含量及Dnmt3a mRNA水平(P<0.05)。0.7g/kg的菟丝子水提液可提高雌鼠Dnmt3amRNA转录水平(P<0.05),1.4g/kg的菟丝子水提液可降低死胎率、BPA水平以及提高E2、P4含量(P<0.01或P<0.05),2.8g/kg的水提液极显著升高子代雌鼠ERα水平(P<0.01)。结果表明,1.4g/kg菟丝子水提液对子宫内BPA暴露小鼠的生殖保护作用较好。
The aim of this study is to explore the reproductive protection effect of Cuscuta chinensis water extract on mice exposed to bisphenol A(BPA)during weaning period.Fifty pregnant female mice were randomly divided into 5groups with 10mice in each group.A model of bisphenol A exposure in the uterus of mice during pregnancy was made and given different doses of Cuscuta.Until the birth of pregnant mice,the pregnancy period(d)and the weight gain(g)of the pregnant mice were recorded.After the offspring mice were born,the total number of pups(only),litter weight(g),average birth weight(g)and stillbirth number(number)were recorded.At 21days of age(weaning),the mice were weighed and blood was collected from the eyeballs.The ratio of male to female and ovarian coefficient were calculated.The ovarian ERαlevels,the content of estradiol(E2),progesterone(P4)and bisphenol A(BPA)in the serum of female offspring were detected by ELISA.mRNA levels of ovarian DNA methyltransferase 3a(Dnmt3a),DNA methyltransferase 3b(Dnmt3b),estrogen receptor alpha(ERα)and progesterone receptor(PR)were detected by realtime quantitative fluorescence PCR(RT-qPCR).The results showed that BPA drinking water could significantly increase the stillbirth rate and serum BPA levels in the offspring of the firstgeneration mice(P<0.05),and significantly reduce the serum E2content and Dnmt3amRNA levels in the offspring of the female mice(P<0.05).0.7g/kg Cuscuta water extract could increase the Dnmt3amRNA transcription level of female mice.1.4g/kg of Cuscuta water extract could reduce stillbirth rate,BPA level and increase E2and P4content(P<0.05or P<0.01).A water extract of 2.8g/kg Cuscuta significantly increased the level of ERαin female offspring(P<0.05).The results indicated that 1.4g/kg Cuscuta water extract has a better reproductive protection effect on mice exposed to intrauterine BPA.
