摘要
通过探究一株来源于健康牦牛粪便的粪肠球菌S-5的安全性及生物学特性,以获得具有治疗犊牦牛春季腹泻病的益生菌群。文章对菌株S-5的革兰氏染色镜检试验、哥伦比亚血平板试验、生化测定试验、耐受性试验、体外抑菌试验、16S rDNA测序鉴定试验,以及动物回归试验的结果进行分析,并验证其安全性及益生性。结果显示,S-5号菌株为无芽孢、无荚膜、无溶血性的阳性球菌,生化测定试验显示其能分解葡萄糖、麦芽糖、乳糖等,产酸不产气,不能分解甘露醇、棉子糖,在硫化氢、硝酸盐、吲哚试验中呈阴性,且血凝试验中不发生凝血反应,耐受性试验显示S-5菌株对牛胆盐浓度为0.03~0.15 g/100 mL时和胰蛋白酶浓度为0.5%~1.3%,pH为4.3~6.5时,耐受性极佳,体外抑菌试验证明S-5菌株对大肠杆菌O111、O78有抑制作用,16S rDNA测序鉴定S-5菌株为粪肠球菌,动物回归试验显示S-5菌株对大肠杆菌O111、O78所致的犊牦牛腹泻病有治疗作用且无毒副作用。结果说明本试验将首次获得一株对大肠杆菌所致犊牦牛春季腹泻病具有治疗效果的西藏牦牛源粪肠球菌S-5,亦符合《微生态制剂的质量鉴别与选择、检测指标和标准》。
In order to obtain a probiotic colony of treating spring diarrhea in calves,by exploring the safety and biological characteristics of a strain Enterococcus faecalis S-5 isolated from healthy yaks'feces.Experi⁃mental results of Gram stain microscopic test,Columbia blood plate test,biochemical test,tolerance test,in vitro antibacterial test,16S rDNA sequencing identification test,and animal regression test,and verified its safety and probiotics were analyzed.The results showed that strain S-5 was a no spore,no capsular,and no hemolytic positive cocci.The result of biochemical determination test showed that it could decompose glu⁃cose,maltose,lactose,etc.,producing acid but no gas,and could not decompose mannitol and cotton seed sugar,which was negative in the hydrogen sulfide,nitrate,and indole tests,and no coagulation reaction oc⁃curred in the hemagglutination test.The results of tolerance test showed that the S-5 strain had anexcellent tolerance,when the concentration of beef bile salt was 0.03~0.15 g/100 mL and the concentration of trypsin was 0.5%~1.3%,pH was 4.3~6.5.In vitro antibacterial test proved that S-5 strain had an inhibitory effect on E.coli O111,E.coli O78,16S rDNA sequencing identified the S-5 strain was Enterococcus faecalis.The animal regression test showed that it had a therapeutic effect on calf diarrhea caused by E.coli O111 and E.coli O78 without toxic side effects.The results indicated that the test would obtain for the first time an En⁃terococcus faecalis from Tibetan yaks,which had a treatment effect to spring diarrhea in calves caused by E.coli,and S-5 was also accorded with the"Qual⁃ity Identification and Selection,Testing Index and Standard of Microecological Preparations".
