摘要
目的探讨BECN1、性别决定区Y盒转录因子9(SOX9)表达与甲状腺癌患者临床特征及预后的关系。方法取106例甲状腺癌患者的甲状腺癌组织及癌旁组织,比较甲状腺癌组织及癌旁组织中BECN1、SOX9表达情况;比较不同临床特征甲状腺癌患者甲状腺癌组织中BECN1、SOX9表达情况;进行为期3年的随访。结果BECN1、SOX9阳性均主要定位于细胞质中。甲状腺癌组织中SOX9阳性表达率明显高于癌旁组织,BECN1阳性表达率明显低于癌旁组织,差异均有统计学意义(P﹤0.01)。Ⅰ~Ⅱ期、高分化、无淋巴结转移的甲状腺癌患者甲状腺癌组织中BECN1阳性表达率分别高于Ⅲ~Ⅳ期、中低分化、有淋巴结转移患者,SOX9阳性表达率分别低于Ⅲ~Ⅳ期、中低分化、有淋巴结转移患者,差异均有统计学意义(P﹤0.05)。BECN1阳性表达患者的平均生存时间为(35.26±8.39)个月,明显长于阴性表达患者的(30.16±7.35)个月;SOX9阳性表达患者的平均生存时间为(31.20±8.03)个月,明显短于阴性表达患者的(35.74±6.99)个月,差异均有统计学意义(P﹤0.01)。多因素Logistic回归模型结果显示,淋巴结转移、分化程度、BECN1及SOX9表达均是甲状腺癌患者预后的影响因素(P﹤0.05)。结论甲状腺癌组织存在BECN1低表达、SOX9高表达,不同临床分期、肿瘤分化程度及淋巴结转移状态患者BECN1及SOX9表达存在差异,且BECN1及SOX9表达与患者预后密切相关。
Objective To investigate the relationship between the expression of BECN1,sex determining region Ybox transcription factor 9(SOX9),the clinical characteristics and prognosis of patients with thyroid carcinoma.Method A total of 106 patients with thyroid cancer were selected,and their thyroid cancer tissues and adjacent tissues were collected to compare the expressions of BECN1 and SOX9 in thyroid cancer tissues and adjacent tissues.The expressions of BECN1 and SOX9 in cancer tissues from thyroid cancer patients with different clinical characteristics were also compared.The duration of the follow-up reached three years.Result Either BECN1 and SOX9 were mainly positive located in the cytoplasm.The positive expression rate of SOX9 in thyroid cancer tissue was significantly higher than that in adjacent tissues,and the positive expression rate of BECN1 was significantly lower than that in adjacent tissues,the differences were statistically significant(P<0.01).The positive expression rate of BECN1 in thyroid cancer tissues of stage I-Ⅱ,well-differentiated,and no lymph node metastasis patients were higher than those in stageⅢ-IV,moderately-differentiated,and lymph node metastasis patients while the positive expression rate of SOX9 were significantly lower than those of patients with poor condition,and the differences were statistically significant(P<0.05).The median survival of patients with positive expression of BECN1 was(35.26±8.39)months,which was significantly longer than that of patients with negative expression(30.16±7.35)months;the median survival of patients with positive expression of SOX9 was(31.20±8.03)months,which was significantly shorter than(35.74±6.99)months of patients with negative expression,the differences were statistically significant(P<0.01).The results of the multivariate Logistic regression model indicated that lymph node metastasis,degree of differentiation,BECN1,and SOX9 expression were all independent risk factors for the prognosis of thyroid cancer patients(P<0.05).Conclusion BECN1 was down-regulation while SOX9 was up-regulation in thyroid carcinoma.The expression of BECN1 and SOX9 varies in patients with different clinical stages,tumor differentiation grade,and lymph node metastasis.The expression of BECN1 and SOX9 is closely related to the prognosis of patients.
