HPLC测定大豆皂苷方法的建立和优化

Establishment and Optimization of HPLC Method for Determination of Soybean Saponins

摘要 为了实现大豆皂苷的快速分离及含量测定,采用高效液相色谱法(HPLC)对4种大豆皂苷(Aa、Ab、Ba、Bb)进行分析检测,建立并优化采用高效液相色谱法(二极管阵列检测器)测定4种大豆皂苷的条件,并采用外标法进行定量分析。结果表明,采用HPLC法测定大豆皂苷的最优条件为:色谱柱ZORBAX SB-C18(250 mm×4.6 mm,5μm),柱温30℃,流动相A为0.05%的乙酸乙腈溶液,流动相B为0.05%的乙酸水溶液,进样30μL,流速1.0 m L/min,检测波长205 nm。大豆皂苷在100~1 000μg/m L范围内线性关系良好,4种大豆皂苷线性相关系数均不低于0.997,大豆皂苷Aa、Ab的检测下限为20μg/m L,Ba、Bb的检测下限为10μg/m L;加标检出率为89.9%~105.6%,相对标准偏差为1.06%~2.31%。该方法操作简单、准确度高、重现性好,可用于大豆种子中4种大豆皂苷的定性定量检测。 To achieve the rapid separation and content determinations of soybean saponins,four kinds of soybean saponins(Aa,Ab,Ba,Bb)were analyzed and detected by high performance liquid chromatography(HPLC).The method for the determination of four kinds of soybean saponins were established and optimized by HPLC with diode array detector,and external standard method was used for quantitative analysis.The result showed that the optimized conditions were a ZORBAX SB-C18(250 mm×4.6 mm,5μm),column temperature was 30℃,mobile phase A was 0.05%in acetonitrile acetate,mobile phase B was 0.05%acetic acid in water,30μL sample was injected,the flow rate was 1.0 mL/min,detection wavelength was 205 nm UV.The soyasaponin had good linearity in the range of 100-1000μg/mL.The correlation coefficients of the four soybean saponin standards(Aa,Ab,Ba,Bb)were not lower than 0.997.The detection limit of soybean saponins Aa,Ab were 20μg/mL,and the detection limit of Ba,Bb were 10μg/mL.The detection rate of spiked standard was 89.9%-105.6%,and the relative deviation(RSD)was 1.06%-2.31%.The method has the advantages of simple operation,short determination time,high accuracy and good reproducibility,and can be used for qualitative and quantitative detection of soybean saponin in soybean seeds.
