超高效液相色谱法同时测定小儿金翘颗粒中10种成分含量

Simultaneous determination of ten constituents in Xiao’er-Jinqiao granules by UPLC

摘要 目的建立超高效液相色谱法同时测定小儿金翘颗粒中绿原酸、葛根素、连翘酯苷A、甘草苷、苦参碱、柴胡皂苷A、柴胡皂苷D、连翘苷、甘草酸铵、靛玉红10种成分含量的方法。方法色谱柱为ACQUITY UPLCTMC18柱(100 mm×2.1 mm,1.7μm);流动相为乙腈-0.05%磷酸水溶液,梯度洗脱,流速0.2 ml/min;检测波长250 nm;柱温30℃;进样量2μl。结果绿原酸、葛根素、连翘酯苷A、甘草苷、苦参碱、柴胡皂苷A、柴胡皂苷D、连翘苷、甘草酸铵、靛玉红的线性范围分别为0.7895~15.7934μg(r=0.9996)、0.5718~11.4456μg(r=0.9994)、0.1107~2.2147μg(r=0.9993)、0.0475~0.9505μg(r=0.9992)、0.2698~5.3958μg(r=0.9997)、0.0851~1.7039μg(r=0.9995)、0.1057~2.1130μg(r=0.9994)、0.0656~1.3121μg(r=0.9993)、0.0805~1.6112μg(r=0.9995)、0.0577~1.1533μg(r=0.9994);定量限分别为8.322、9.023、9.857、4.583、6.872、7.844、8.643、5.895、7.568、5.251μg/ml;平均加样回收率分别为99.11%、98.87%、97.83%、98.10%、98.17%、98.30%、98.10%、98.14%、97.99%、98.02%(RSD<2.0%);精密度、重复性、稳定性试验的RSD<2.0%。结论所建立的多成分含量测定方法快捷、准确、重复性好,可用于小儿金翘颗粒的质量控制。 Objective To establish a method for simultaneously determinating the content of 10 constituents including chlorogenic acid,puerarin,forsythoside A,liquiritin,matrine,saikosaponin A,saikosaponin D,forsythin,ammonium glycyrrhizate,indirubin in Xiao’er-Jinqiao granules by UPLC.Methods ACQUITY UPLCTMC18 chromatographic column(100 mm×2.1 mm,1.7μm)was used.The mobile phase was acetonitrile-0.05%phosphoric acid solution with gradient elution at the flow rate of 0.2 ml/min.The detection wavelength was 250 nm.The column temperature was 30℃.The injection volume was 2μl.Results The linear ranges of chlorogenic acid,puerarin,forsythoside A,liquiritin,matrine,saikosaponin A,saikosaponin D,forsythin,ammonium glycyrrhizate,indirubin were 0.7895-15.7934μg(r=0.9996),0.5718-11.4456μg(r=0.9994),0.1107-2.2147μg(r=0.9993),0.0475-0.9505μg(r=0.9992),0.2698-5.3958μg(r=0.9997),0.0851-1.7039μg(r=0.9995),0.1057-2.1130μg(r=0.9994),0.0656-1.3121μg(r=0.9993),0.0805-1.6112μg(r=0.9995),0.0577-1.1533μg(r=0.9994).The limits of quantitation were 8.322,9.023,9.857,4.583,6.872,7.844,8.643,5.895,7.568,5.251μg/ml.The average recovery rates were 99.11%,98.87%,97.83%,98.10%,98.17%,98.30%,98.10%,98.14%,97.99%and 98.02%,and RSDs were all below 2.0%,respectively.RSDs of precision,reproducibility and stability tests were lower than 2.0%.Conclusions The established method of multi-component content determination is fast,accurate and reproducible.It can be used for the quality control of Xiao’er-Jinqiao granules.
