青蒿治疗系统性红斑狼疮的网络药理学分析 被引量:3

摘要 目的:通过网络药理学方法研究青蒿治疗系统性红斑狼疮(SLE)的潜在作用机制。方法:通过中国传统药理学数据库和分析平台(TCMSP)筛选青蒿的有效活性成分和作用靶点,运用Uniprot数据库将药物靶点进行基因名字标准化处理,在GeneCards、OMIM数据库中检索SLE相关靶点,使用R软件绘制Venny图找出青蒿与SLE共同作用靶点,String数据库构建蛋白质互作(PPI)网络,Cytoscape软件中进行PPI网络可视化,构建“药物-活性成分-潜在靶点-疾病”网络,并筛选出核心基因,对共同作用靶基因进行GO、KEGG富集分析。结果:筛选得到青蒿的有效活性成分22个,111个青蒿有效成分与SLE共同作用靶基因,30个核心基因。GO功能富集显示青蒿治疗SLE生物学过程和功能主要集中在细胞因子受体结合、细胞因子活性、核受体活性、转录因子活性、受体配体活性、类固醇激素受体等。KEGG通路富集显示,青蒿治疗SLE主要涉及TNF、IL-17、Toll样受体、Th17细胞分化等信号通路。结论:通过构建“药物-活性成分-潜在靶点-疾病”网络,阐释了青蒿多成分、多靶点、多条通路治疗SLE的作用机制,为深入研究青蒿治疗SLE的机制提供一定参考意义。 Objective:To study the potential mechanism of Artemisia annua in the treatment of systemic lupus erythematosus(SLE)by means of network pharmacology.Method:Screen the effective active ingredients and targets of Artemisia annua L.through the Chinese Traditional Pharmacology Database and Analysis Platform(TCMSP),use the Uniprot database to standardize the gene names of the drug targets,and search for SLE-related targets in the Gene-Cards and OMIM databases,Use R software to draw Venny diagram to find out the co-action target of Artemisia annua and SLE,String database to construct protein interaction(PPI)network,Cytoscape software to visualize PPI network,construct“drug-active ingredient-potential target-disease”Network,and screen out the core genes,and perform GO and KEGG enrichment analysis on the co-acting target genes.Results:22 effective active ingredients of A.annua were screened,111 active ingredients of A.annua and SLE co-act target genes,and 30 core genes.GO function enrichment shows that the biological processes and functions of Artemisia annua in the treatment of SLE mainly focus on cytokine receptor binding,cytokine activity,nuclear receptor activity,transcription factor activity,receptor ligand activity,and steroid hormone receptor.Enrichment of KEGG pathway shows that the treatment of SLE by Artemisia annua mainly involves signal pathways such as TNF,IL-17,Toll-like receptors,and Th17 cell differentiation.Conclusion:By constructing a“drug-active ingredient-potential target-disease”network,the mechanism of multi-component,multitarget,and multiple pathways of Artemisia annua L.in the treatment of SLE is explained,and it provides a reference for in-depth study of the mechanism of Artemisia annua in the treatment of SLE.significance.
