磁性壳聚糖微球固定化NAD激酶的制备及稳定性 被引量:1

摘要 探究NAD激酶固定化条件及固定化酶的相关稳定性。制备磁性壳聚糖微球,戊二醛做交联剂把NAD激酶固定在磁性壳聚糖微球上。对影响固定化酶酶活的因素(温度、交联时间、pH及戊二醛浓度)进行考察,并设计正交试验对固定化条件进行优化。研究固定化酶的操作稳定性及储存稳定性。酶固定化最优条件为固定温度60℃、p H 9.0、戊二醛体积分数3.75%、交联时间6 h,该条件下酶活力回收率为56.16%,验证试验表明该工艺制备稳定;固定化酶在重复使用8次后,固定化酶酶活力回收率剩余55.89%;固定化酶储存10 d后酶活力回收率为93.12%。成功制备了固定化酶,固定化酶稳定性较好。 To investigate the immobilization conditions of NAD kinase and the related stability of immobilized enzymes.Magnetic chitosan microspheres were prepared and glutaraldehyde was used as a cross-linking agent to immobilize NAD kinase on magnetic chitosan microspheres.The factors affecting the activity of immobilized enzymes(temperature,crosslinking time,pH and glutaraldehyde concentration)were investigated,and orthogonal experiments were designed to optimize the immobilization conditions.The operational stability and storage stability of the immobilized enzyme were investigated.The optimal conditions for enzyme immobilization were fixed temperature 60℃,pH 9.0,glutaraldehyde volume fraction 3.75%,cross-linking time 6 h,and the enzyme activity recovery rate was 56.16%under this condition.The verification test showed that the preparation was stable;After the enzyme was used for 8 times,the recovery rate of the immobilized enzyme activity was 55.89%.The enzyme activity recovery rate after immobilized enzyme storage for 10 d was 93.12%.The immobilized enzyme was successfully prepared,and the immobilized enzyme had good stability.
