摘要
目的采用蛋白质组学方法探讨黄芩黄连药对改善2型糖尿病KK-Ay小鼠肝组织的炎症作用机制。方法从2型糖尿病模型组和黄芩黄连药对组KK-Ay小鼠肝组织中提取全蛋白,使用nano-LC-LTQ/Orbitrap MS检测差异蛋白,使用Proteome Discoverer软件鉴定蛋白,Sieve软件对肝组织蛋白进行数据分析处理,并通过DAVID数据库KEGG pathway、String等生物信息学方法筛选相关差异蛋白。结果经蛋白质组学研究共发现,模型组与黄芩黄连药对组间存在与肝组织炎症相关的差异蛋白,包括TRAF6(肿瘤坏死因子受体相关因子6)、IL-1α(白细胞介素1α)、NF-κB2(核因子-κB2)、RPS6KA1(核糖体蛋白S6激酶α1)等。经DAVID和KEGG通路分析,这些差异蛋白主要参与炎症相关信号通路。结论黄芩黄连药对改善2型糖尿病KK-ay小鼠肝组织炎症作用与TRAF6、IL-1α、NF-κB2、RSK1、RSK2蛋白表达下调相关。该研究为黄芩黄连药对临床用于防治代谢性疾病提供了实验依据。
Objective To explore the mechanism of Scutellaria-coptis herb couple on improving the inflammation of liver tissue in type 2 diabetic KK-Ay mice by proteomics. Methods Whole protein was extracted from the liver tissue of KK-Ay mice of type 2 diabetes model group and Scutellaria-coptis herb couple group, and the differential protein was detected by nano-LC-LTQ/Orbitrap MS. Proteome Discoverer software was used to identify the protein. The tissue proteins were analyzed and processed, and related differential proteins were screened by bioinformatics methods such as DAVID database KEGG pathway and String. Results Through proteomics research, it was found that there were differential proteins related to liver tissue inflammation between the model group and the Scutellaria-coptis herb couple group, including TRAF6(tumor necrosis factor receptor-related factor 6), IL1α(interleukin 1α), NF-κB2(nuclear factor-κB2), RPS6 KA1(ribosomal protein S6 kinase α1), etc. After analyzing the DAVID and KEGG pathways, these differential proteins were closely related to inflammation. Conclusion the effect of Scutellaria-coptis herb couple on improving inflammation of liver tissue in KK ay mice with type 2 diabetes mellitus is related to the down-regulation of TRAF6, IL1 α, NF-κ B2, rSK1 and RSK2 protein expression, which provides experimental basis for Scutellaria-coptis herb couple to prevent and treat metabolic diseases in clinic.
