摘要
目的观察岭南火针对带状疱疹后神经痛(postherpetic neuralgia,PHN)大鼠行为学和背根神经节(dorsal root ganglion,DRG)中蛋白激酶A(PKA)、香草酸瞬时受体亚型1(TRPV1)、磷酸化香草酸瞬时受体亚型1(pTRPV1)的影响,探讨火针干预PHN的镇痛机制。方法SD大鼠随机分为正常组(Control)、模型组(RTX)、溶剂组(Solvent)、火针组(FA)和阳性对照组(GBP),每组10只。采用树脂毒素(RTX)腹腔注射制备PHN模型,观察并记录每组疼痛行为学机械缩足阈值(PWT)、热痛阈值(TWL);Western blot检测DRG中PKA、TRPV1、pTRPV1的蛋白表达。结果与Control组相比,造模后,PWT显著降低,TWL显著升高(P<0.01)。第12天,FA组、GBP组PWT值明显高于RTX组(P<0.05)。第16天,FA组、GBP组TWL值明显低于RTX组(P<0.05)。第22天,FA组、GBP组的PWT、TWL值差异有统计学意义(P<0.05)。与Control组相比,RTX组的PKA、TRPV1、pTRPV1蛋白表达明显降低(P<0.05),FA组、GBP组的各蛋白水平较RTX组明显升高(P<0.01),FA组和GBP组组间各蛋白表达水平相比差异无统计学意义(P>0.05)。结论火针能有效地缓解PHN大鼠的周围神经痛,并激活PKA-TRPV1通路,其镇痛机制可能与有效地调控背根神经节中PKA、TRPV1、pTRPV1蛋白的表达有关;其镇痛作用的发挥可能是通过该通路实现的。
Objective To observe the effects of Lingnan fire needle on PKA,TRPV1 and pTRPV1 in post herpetic neuralgia(PHN)rats′behavior and dorsal root ganglion(DRG),and to explore the intervention of fire needle on PHN analgesic mechanism.Methods SD rats were randomly divided into normal group(Control),model group(RTX),solvent group(Solvent),fire needle group(FA)and positive control group(GBP),with 10 rats in each group.The PHN model was prepared by intraperitoneal injection of resin toxin(RTX).Observe and record the mechanical foot withdrawal threshold(PWT)and thermal pain threshold(TWL)of each group of pain behavior;WB detected the protein expression of protein kinase A(PKA),vanillic acid transient receptor subtype 1(TRPV1),phosphorylated vanillic acid transient receptor subtype 1(pTRPV1)in DRG.Results Compared with the control group,PWT was significantly reduced after modeling,and TWL was significantly increased(P<0.01).On the 12 th day,the PWT of FA group and GBP group was significantly higher than that of RTX group(P<0.05).On the 16 th day,the TWL value of FA and GBP group was significantly lower than that of RTX group(P<0.05).On the 22 th day,the PWT and TWL values of the FA and GBP groups were significantly different and statistically significant(P<0.05).Compared with the control group,the protein expressions of PKA,TRPV1 and pTRPV1 in the RTX group were significantly reduced(P<0.05),and the protein levels of the FA and GBP group were significantly higher than those in the RTX group(P<0.01).The FA and GBP groups conclusion of protein expression levels between groups(P>0.05).Conclusion Fire needle can effectively relieve peripheral neuralgia in PHN rats and activate the PKA-TRPV1 pathway.Its analgesic mechanism may be related to the effective regulation of the expression of PKA,TRPV1,and pTRPV1 proteins in the dorsal root ganglia.Its analgesic effect may be achieved through this pathway.
