摘要
目的分析枸杞多糖(LBP)对小鼠肝纤维化组织中基质金属蛋白酶-9(MMP-9)及其抑制剂-1(TIMP-1)含量的影响,探讨LBP对肝纤维化的作用机制。方法胆总管结扎方法制造肝组织纤维化损伤小鼠,将12只小鼠按照随机数字法分组为肝纤维化组和LBP组,同时选取6只正常小鼠作为正常组。LBP组:肝组织纤维化小鼠每天经口服用LBP 25μg/g;正常组及肝纤维化组:经口服用等体质量的生理盐水,3组同时连续服用3周。取出各组的肝组织,分别经苏木精-伊红染色、天狼猩红染料染色后,观察其结构改变情况以及肝组织中胶原纤维的含量;蛋白印迹技术评估肝组织中MMP-9和TIMP-1的含量。结果肝纤维化组的肝组织结构紊乱,其内有程度不同的坏死组织,红色的胶原纤维面积和汇管区域小叶间胆管数量比正常组增多,肝组织内MMP-9的含量(0.044±0.011)比正常组增加,但是两组相比差异无统计学意义(P>0.05),TIMP-1的含量(0.712±0.038)比正常组增加(P<0.05),MMP-9/TIMP-1的值(0.051±0.011)比正常组减少(P<0.05)。LBP组肝组织的坏死程度减轻,红色的胶原纤维面积和汇管区域小叶间胆管数量比肝纤维化组减少,肝组织内MMP-9的含量(0.070±0.009)比肝纤维化组增多(P<0.05),TIMP-1的含量(0.445±0.027)比肝纤维化组减少(P<0.05),MMP-9/TIMP-1的值(0.163±0.025)比肝纤维化组增加(P<0.05)。结论LBP能同时调节肝纤维化组织中MMP-9和TIMP-1的含量,使MMP-9/TIMP-1的值增加来有效阻止肝组织纤维化的进展,达到保护肝组织的目的。
Objective To analyze the effect of lycium barbarum polysaccharide(LBP)on the content of matrix metalloproteinase-9(MMP-9)and its inhibitor-1(TIMP-1)in hepatic fibrosis tissue of mouse,explore the mechanism of LBP on liver fibrosis.Methods Mice with hepatic tissue fibrosis injury were made through complete obstruction of common bile duct.12 mice were arranged into hepatic fibrosis group and LBP group according to random number method.Meanwhile,6 normal mice were selected as normal group.LBP group:mice with liver fibrosis were given LBP 25μg/g by oral administration every day;normal group and liver fibrosis group:mice were given normal saline of equal weight by oral administration.Three groups were treated for three weeks.The liver tissues of each group were stained with hematoxylin eosin and Sirius red respectively,the changes of liver structure and the content of collagen fiber in hepatic tissue were observed,and the contents of MMP-9 and TIMP-1 in hepatic tissue were evaluated by Western blot.Results In the fibrosis group,the structure of liver tissue was disordered,there were necrosis tissues in different degrees,the area of red collagen fiber and the number of interlobular bile ducts in the manifold area were more than those in the normal group,and the content of MMP-9(0.044±0.011)in liver tissue was more than its in normal group,but the difference was not statistically significant(P>0.05),TIMP-1 content(0.712±0.038)was higher than that in normal group(P<0.05),and the value of MMP-9/TIMP-1(0.051±0.011)was lower than that in normal group(P<0.05).In LBP group,the necrosis degree of liver tissue decreased,the area of red collagen fiber and the number of interlobular bile ducts in the portal area decreased as compared with the hepatic fibrosis group,the content of MMP-9 in liver tissue(0.070±0.009)was higher than that in the hepatic fibrosis group(P<0.05),TIMP-1 content(0.445±0.027)was lower than that in the hepatic fibrosis group(P<0.05),and the value of MMP-9/TIMP-1(0.163±0.025)was higher than that in the hepatic fibrosis group(P<0.05).Conclusion LBP can simultaneously regulate the content of MMP-9 and TIMP-1 in liver fibrosis tissue,increase the value of MMP-9/TIMP-1 to effectively prevent the progress of liver fibrosis,and achieve the purpose of protecting liver tissue.
