摘要
为明确向日葵核盘菌(Sclerotinia sclerotiorum)菌核的菌丝型萌发特性,采用控制变量法测定不同因素对菌核菌丝型萌发的影响。结果表明,温度为5~30℃时菌核均能进行菌丝型萌发,最适温度为25℃,30℃时菌核菌丝型萌发后无法形成菌核;酸性条件利于菌核进行菌丝型萌发和菌核形成;光照时间越长越利于菌核菌丝型萌发及萌发后形成菌核,而紫外光照射2 h对其萌发和菌核的形成无明显抑制作用;菌核的菌丝型萌发最佳碳源为蔗糖,氮源为蛋白胨,形成菌核最佳碳源为乳糖,氮源为硝酸铵;菌核致死温度为70℃处理10 min。
In order to clarify the myceliogenic germination characteristics of sclerotia of Sclerotinia sclerotiorum,the influencing factors of myceliogenic germination were studied by control variable method.The results showed that sclerotia could germinate hypha at 5-30℃,the most favorable temperature for sclerotia germination was 25℃,and sclerotia could not beformed after hyphal germination at 30℃.Acidic conditions were favorable for myceliogenic germination and sclerotia formation.The longer the illumination time was,the faster the hyphae sprouting from the sclerotia grew and the more sclerotia were formed.The ultraviolet irradiation had no obvious inhibitory effect on the mycelial germination and sclerotial production.The optimum carbon source for mycelial germination of sclerotia was sucrose and nitrogen source was peptone.The optimum carbon source for sclerotia formation was lactose and nitrogen source was ammonium nitrate.The lethal temperature for sclerotia was 70℃for 10 minutes.
