莱菔子配伍蒺藜对自发性高血压大鼠的降压作用及机制研究 被引量:8

摘要 目的:探讨莱菔子配伍蒺藜对自发性高血压大鼠(SHR)血压、胸主动脉和心脏组织的影响。方法:40只SHR大鼠随机分为模型组、莱菔子组、蒺藜组、莱菔子配伍蒺藜(莱蒺)组、马来酸依那普利组,8只同周龄WKY大鼠作为空白对照组。连续给药4周,每周测量大鼠尾动脉收缩压,末次给药后观察大鼠胸主动脉和心肌组织的病理学变化,ELISA法测定血清中内皮素-1(ET-1)、血管紧张素(AngⅡ)、醛固酮(ALD)和NO的表达。结果:与模型组比较,莱菔子组、蒺藜组、莱菔子配伍蒺藜组能够降低SHR大鼠收缩压(P<0.01)以及大鼠血清中ET-1、AngⅡ、ALD含量,升高血清中NO含量(P<0.05或P<0.01),降低血管壁的厚度,抑制胸主动脉血管重构,改善心肌细胞形态。结论:莱菔子配伍蒺藜能干预血压的升高,对胸主动脉和心肌组织有保护作用,其机制可能与降低血清中ET-1、AngⅡ、ALD含量、增加血清中NO含量有关。 Objective:To explore the effects of Semen raphani combined with Tribuli fructus on blood pressure,thoracic aorta and heart tissue in spontaneous hypertension rats(SHR).Methods:40 SHR were randomly divided into Semen raphani group,Tribuli fructus group,Semen raphani combined with tribuli fructus group,enalapril group,and 8 Wistar-Kyoto(WKY)rats of the same age as the normal control group.Systolic pressure of the caudal artery were measured every week.Four weeks after the administration,the levels of ET-1,AngⅡ,ALD and NO in the blood serum were measured.Pathological changes of thoracic aorta and heart tissue were observed by HE staining.Results:The systolic blood pressure in experimental groups were significantly lower than those in model group(P<0.01),the levels of ET-1,AngⅡ,ALD were also significantly decreased,but the levels of NO were significantly increased and the difference was statistically significant(P<0.05 or P<0.01).The experimental groups could significantly reduce the thickness of the vessel wall and inhibit the reconstruction of thoracic aortic vascular,improve the pathological changes such as irregular cell morphology,proliferation of fibrous tissue and so on.Conclusion:Semen raphani combined with tribuli fructus had a protective effect on the thoracic aorta and heart tissue and could interfere with the increase of blood pressure,which might be related to reducing serum ET-1,AngⅡ,ALD content and increasing the content of NO in serum.
