摘要
目的探究CD44、对醌氧化还原酶(NQO1)基因多态性与肺癌易感性及化疗预后的相关性。方法选取98例肺癌患者(肺癌组)和100例健康对照人群(对照组),选择CD44基因的rs8193和rs13347两个SNP位点,并选择NQO1基因rs1437135、rs10517和rs18005663个位点进行基因多态性检测,分析2组分布特征;分析CD44、NQO1基因多态性与肺癌患者临床病理特征及化疗疗效的关系。结果CD44基因的rs8193、rs13347基因位点及NQO1基因rs1437135、rs10517、rs1800566基因位点的基因型和分布频率在肺癌组及对照组比较差异有统计学意义(P<0.05)。对比化疗显效和无效肺癌患者性别、年龄、吸烟史、病理类型差异无统计学意义(P>0.05);化疗显效和无效肺癌患者CD44基因的rs8193、rs13347基因位点及NQO1基因rs1437135、rs10517、rs1800566基因位点多态性比较差异具有统计学意义(P<0.05)。不同CD44、NQO1基因型的肺癌患者中位生存时间(MST)比较差异具有统计学意义(P<0.05)。结论CD44、NQO1基因多态性与肺癌易感性、化疗效果及远期生存存在密切关系。CD44基因rs8193位点CC基因型、NQO1基因rs1437135位点AA基因型及rs1800566位点CC型可能为抑制肺癌发生的保护因素,CD44基因rs13347位点CC基因型、NQO1基因rs10517位点CC基因型为肺癌发生的风险因素。
Objective To explore the correlation between CD44 and NAD(P)H quinone oxidoreductase 1(NQO1)gene polymorphisms and lung cancer susceptibility and prognosis of chemotherapy.Methods Gene polymorphism detection was performed in 98lung cancer patients(lung cancer group)and 100 healthy controls(control group)includingr s8193 and rs13347 of CD44 gene and rs1437135,rs10517 and rs1800566 of NQO1 gene.The relationship between gene polymorphisms of CD44 and NQO1 and clinicopathological characteristics of lung cancer patients and chemotherapeutic effect were analyzed.ResultsThe genotype and distribution frequency of the rs8193,rs13347 locus of CD44 gene and the rs1437135,rs10517,rs1800566 locus of NQO1 gene were significantly different between the lung cancer group and the control group(P<0.05).There was no statistically significant difference in gender,age,smoking history,pathological type of lung cancer patients with markedly effective and ineffective chemotherapy(P>0.05).The differencesof gene locus polymorphismswere statistically significant in rs8193,rs13347 gene loci of CD44 gene and rs1437135,rs10517,rs1800566 of NQO1 genes in lung cancer patients with markedly effective and ineffective chemotherapy(P<0.05).The median survival time(MST)of the lung cancerpatients with different CD44,NQO1 genotypes was significantly different(P<0.05).Conclusions CD44 and NQO1 gene polymorphisms related to lung cancer susceptibility,chemotherapy effect and long-term survivalclosely.The CC genotype at the rs8193 locus of the CD44 gene,the AA genotype at the rs1437135 locus of the NQO1 gene,and the CC type at the rs1800566 locus of the CD44 gene may be protective factors for suppressing lung cancer.The CC genotype at the rs13347 locus of the CD44 gene and the CC genotype at the rs10517 locus of the NQO1 gene are risk factors for lung cancer.
