民族药六棱菊抗炎作用及其机制研究 被引量:8

摘要 目的:研究六棱菊提取物的抗炎作用及其机制,为该药用植物开发应用提供实验依据。方法:建立二甲苯致小鼠耳肿胀、醋酸致小鼠腹腔毛细血管通透性增加、角叉菜胶致小鼠足肿胀等炎症模型,评价六棱菊提取物对急、慢性炎症的药理活性。建立二甲苯致摘除双侧肾上腺小鼠耳肿胀、角叉菜胶致摘除双侧肾上腺小鼠足肿胀模型,判断其抗炎作用是否依赖下丘脑-垂体-肾上腺轴(HPAA)系统,测定足炎性组织丙二醛(MDA)、一氧化氮(NO)和超氧化物歧化酶(SOD)水平。结果:与模型组比较,除低剂量组无统计学意义(P>0.05)外,六棱菊提取物高、中剂量组对二甲苯致小鼠耳肿胀均有显著的抑制作用;六棱菊提取物抑制醋酸致小鼠腹腔毛细血管通透性的升高,对角叉菜胶致小鼠足肿胀亦有显著抑制作用;与模型组比较,在摘除双侧肾上腺小鼠模型中,六棱菊提取物高、中剂量组能显著抑制二甲苯致该模型小鼠耳肿胀;以及能显著抑制角叉菜胶致该模型小鼠足肿胀;六棱菊提取物高、中剂量组均能显著降低足组织中MDA和NO水平,均能显著提高足组织中SOD水平。结论:六棱菊提取物均具有抗炎活性,其抗炎作用的发挥不依赖HPAA系统,同时六棱菊提取物的抗炎作用机制与降低MDA和NO水平,提高SOD水平有关。 Objective:To study the anti-inflammatory effect and its mechanism of Laggera alata extract,and to provide experimental basis for the development and application of the medicinal plant.Methods:Inflammatory models were established,including ear swelling in mice,increased peritoneal capillary permeability induced by glacial acetic acid,and swelling of the footpad in mice caused by carrageenan.The pharmacological activity of Laggera alata extract on acute and chronic inflammation was evaluated.Xylene-induced swelling and carrageenan-induced swelling models in bilateral adrenal gland removed mice were established to determine whether the anti-inflammatory effect of Laggera alata extract depends on the hypothalamic pituitary adrenal axis(HPAA)system.And the levels of MDA,NO,SOD in inflammatory tissue of footpad were determined.Results:Compared with blank group,there was no statistical significance(P>0.05)except for low-dose group.The high-and medium-dose of Laggera alata extract had significant inhibitory effect in swelling mice model;the high-,medium-and low-dose groups significantly increased the peritoneal capillary permeability in mice induced by glacial acetic acid,and the carrageenan-induced swelling in mouse footpads was significantly inhibited.Compared with blank group,the bilateral adrenal gland removed group showed significantly decreased ear swelling induced by xylene,and decrease footpad swelling induced by carrageenan in high and medium doses of Laggera alata extract.The levels of MDA and NO in toes were significantly reduced,and the SOD level in the footpads was significantly increased in both high and medium dose groups.Conclusion:All doses of Laggera alata extract had anti-inflammatory activity,and their anti-inflammatory effects are independent of hypothalamic pituitary adrenal axis(HPAA).At the same time,the anti-inflammatory mechanism of the extract of Laggera alata was associated with reduced levels of MDA and NO and increased level of SOD.
