摘要
本文采用高效液相色谱-柱后衍生法对变性淀粉中的黄曲霉毒素G_(2)、G_(1)、B_(2)和B_(1)含量进行测定。样品采用乙腈-水(84∶16,v/v)提取,经免疫亲和柱净化,C18色谱柱分离,以乙腈-甲醇-水(10∶30∶60,v/v)为流动相进行洗脱,用HPLC光化学衍生-荧光检测器检测样品含量。结果表明,4种黄曲霉毒素的标准曲线线性均大于0.9993,回收率96%~104%,黄曲霉G_(2)、G_(1)、B_(2)和B_(1)的检出限分别为0.0100μg/kg、0.0300μg/kg、0.0100μg/kg和0.0300μg/kg。该方法操作简便,准确度高,适用于变性淀粉中黄曲霉毒素的测定。与柱前衍生法相比,该法能节省检测时间和检测成本,同时减少化学试剂对检测人员的伤害。
In this paper,the contents of aflatoxin G_(2)、G_(1)、B_(2)、B_(1) in modified starch were determined by high performance liquid chromatography(HPLC)and post-column photochemical reaction.The samples were extracted by acetonitrile-water(84∶16,v/v),purified by immunoaffinity column,separated by C_(18) column,eluted with acetonitrile-methanol-water(10∶30∶60,v/v)as mobile phase,and then the contents of samples were detected by post-column photochemical derivation-fluorescence detector.The results showed that the correlation coefficients of all aflatoxins were more than 0.9993,the recoveries were 96%~104%,and the detection limits of aflatoxin G_(2)、G_(1)、B_(2)、B_(1) were 0.0100μg/kg,0.0300μg/kg,0.0100μg/kg and 0.0300μg/kg,respectively.The method was simple,accurate and suitable for the determination of aflatoxin in modified starch.Compared with the pre-column derivatization method,this method could save the detection time and cost,and at the same time,reduce the harm of the chemical reagents to the testers.
