高效液相色谱法测定甘草流浸膏中赭曲霉毒素A的含量 被引量:1

摘要 目的建立甘草流浸膏中赭曲霉毒素A的免疫亲和层析净化高效液相色谱法含量测定方法。方法采用HypersilBDC-C18色谱柱(250.0 mm×4.6 mm,5μm),以乙腈-2%冰醋酸(48∶52)为流动相,激发波长为333μm,发射波长为460μm,流速1.0 ml/min,柱温35℃,进样量10μl,对赭曲霉毒素A的测定。结果在上述色谱条件下,对赭曲霉毒素和相邻其他色谱峰之间分离度良好。赭曲霉毒素A线性回归方程为Y=221.23X+170.72(r=0.9987);在1.0~50.0μg/ml时,赭曲霉毒素A的质量浓度与峰面积呈现良好的线性关系,平均回收率为92.48%。结论该方法操作简单、方便、可靠,可为甘草流浸膏等中成药中真菌毒素的质量控制提供借鉴。 Objective To establish an immunoaffinity chromatography method in the determination of ochratoxin A in licorice root extract by high performance liquid chromatography.Methods The Hypersil BDC-C18 chromatographic column(250.0 mm×4.6 mm,5μm)was used to determined ochratoxin A under the conditions as below:acetonitrile-2%glacial acetic acid(48∶52)was the mobile phase,the excitation wavelength was 333μm,the emission wavelength was 460μm,the flow rate was 1.0 ml/min,the column temperature was 35℃,and the injection volume was 10μl.Results The separation between ochratoxin and other adjacent chromatographic peaks was good.The linear regression equation of ochratoxin A was Y=221.23X+170.72(r=0.9987).There was a good linear relationship when the ochratoxin A was 1.0-50.0 μg /ml, and the average recovery rate was 92.48%. Conclusion This method is simple, convenient, and reliable, and can be used for the quality control of ochratoxin A in licorice root extract and other proprietary Chinese medicine.
