摘要
目的:通过观察夹脊电针对肌萎缩侧索硬化症转基因小鼠(ALS-SOD1G93A)腰髓运动神经元细胞凋亡及Bax、Bcl-2、Caspase-3蛋白表达的影响,探讨夹脊电针防治ALS的作用机制。方法:将36只雄性ALS-SOD1G93A小鼠随机分为模型组、手针组和电针组,每组12只,同窝12只雄性野生型小鼠作为野生组。手针组针刺腰部夹脊穴,每周2次,每次20min,连续治疗4周;电针组采用电针腰部夹脊穴治疗,电针刺激量为1mA,2Hz,疗程同手针组。转棒实验检测各组小鼠运动功能,TUNEL染色观察各组小鼠腰髓前角运动神经元细胞凋亡情况,免疫组织化学及Western Blot法检测小鼠腰髓Bax、Bcl-2、Caspase-3表达水平。结果:与野生组比较,模型组转棒时间下降(P<0.05),腰髓前角凋亡细胞增加,Bax、Caspase-3阳性细胞增加,Bcl-2阳性细胞减少,腰髓组织Bax、Caspase-3表达水平升高,Bcl-2表达水平降低(P<0.01);与模型组比较,手针组、电针组转棒时间显著延长(P<0.05),腰髓前角凋亡细胞减少,Bax、Caspase-3阳性细减少,Bcl-2阳性细胞增加,腰髓组织Bax、Caspase-3表达水平明显降低,Bcl-2表达水平明显升高,且电针组改善程度更为显著(P<0.01)。结论:夹脊电针可能通过调节ALS-SOD1G93A小鼠腰髓组织Bax、Bcl-2、Caspase-3蛋白表达,影响运动神经元细胞凋亡,从而延缓ALS的进展。
Objective: To observe the effect of Jiaji electro-acupuncture on the apoptosis of lumbar spinal cord motor neurons and the expression of Bax, Bcl-2 and Caspase-3 proteins for ALS-SOD1G93 A amyotrophic lateral sclerosis transgenic mice, discuss the mechanism of Jiaji electro-acupuncture in preventing and treating ALS. Methods: 36 male ALS-SOD1G93 A mice were randomly divided into model group, acupuncture group and electro-acupuncture group, with 12 mice in each group, 12 male wild-type mice in the same litter were used as the wide-type group. In the acupuncture group, the lumbar Jiaji point was acupunctured twice a week for 20 min each time, and the treatment lasted for four weeks. In the electro-acupuncture group, eleco-acupuncture at Jiaji points was performed, the stimulation amount was set as 1 mA, 2 Hz, and the course of treatment was the same as that of the acupuncture group. The motor ability of the mice in each group was detected by the rotarod test, the apoptosis of the motor neurons in the lumbar ventricornu of the mice in each group was detected by TUNEL staining, and the expression of Bax, Bcl-2 and Caspase-3 proteins in mouse lumbar spinal cord was detected by immunohistochemistry and Western Blot. Results: Compared with the wild-type group, the rotarod time in the model group were decreased(P < 0.05), the apoptotic cells in the lambar ventricornu increased, the Bax and Caspase-3 positive cells increased, the Bcl-2 positive cells decreased, the expression levels of Bax and Caspase-3 in the lumbar spinal cord tissue increased, and the expression levels of Bcl-2 decreased(P < 0.01). Compared with the model group, the rotarod time of the acupuncture group and the electro-acupuncture group was significantly prolonged(P < 0.05), the apoptotic cells of the lumbar ventricornu were decreased, the positive Bax and Caspase-3 cells were decreased, the positive Bcl-2 cells were increased, the expression levels of Bax and Caspase-3 in the lumbar spinal cord tissues were significantly decreased, the expression level of Bcl-2 was significantly increased, and the improvement of the electro-acupuncture group was more significant(P < 0.01). Conclusion: The Jiaji electro-acupuncture may affect the apoptosis of motor neurons by regulating the expression of Bax, Bcl-2 and Caspase-3 proteins in the lumbar spinal cord of in ALS-SOD1G93 A mice, thereby delaying the progression of ALS.
